Project description:Trout spermatogenesis proceeds seasonally in a way that all morphological and cellular events tend to be synchronized. We thus used gonads at key stages of the male reproductive cycle together with isolated germ cell populations to study the changes in gene expression underlying testis development. Statistical analysis followed by clustering of expression profiles allowed the discrimination of sequential events of gene activation or repression during testis development and/or throughout the germline. 38 samples covering 6 developmental stages (as defined in Gomez et al. 1998 Biol. Reprod. 58, 483-491) and 3 isolated germ cell populations were analysed. This included: - Testes at early stages containing slowly-dividing type A spermatogonia (Stage_I, n=5) or growing numbers of actively-dividing type B spermatogonia (Stages_IIa, n= 4; Stage_IIb, n= 4) - Maturing testes containing in addition large numbers of meiotic spermatocytes (Stage_IIIb, n=3) and post-meiotic spermatids (Stage_V, n=4) - Spawning testes containing essentially mature spermatozoa (Stage_VIII, n=3) - Fractions of isolated germ cells enriched in spermatogonia (Spermatogonia, N=6), spermatocytes (Spermatocyte, N=6) and spermatids (Spermatid, N=3).

Project description:We show that Glis3 is expressed in gonocytes, SSCs and SPCs, but not in differentiated spermatogonia or subsequent stages of spermatogenesis nor in Sertoli or Leydig cells. We further demonstrate that Glis3-deficiency causes a severe impairment in spermatogenesis in mice. Although the number of gonocytes was slightly diminished in Glis3KO testis, the number undifferentiated, PLZF+ spermatogonia was dramatically reduced leading to a virtual block in the progression of spermatogenesis. Gene expression profiling showed that the expression of a number of genes associated with self-renewal and differentiation of spermatogonial cells was significantly decreased in 1-week-old Glis3KO2 testis. These included a set of GDNF-dependent genes, such as Etv5, Bcl6b, Lhx1, Brachyury, Id4, and Pou3f1, and GDNF-independent genes, such as FoxO1, Oct4, and Zbtb16. Impairment of the nuclear localization of FoxO1 may be in part responsible for the reduced expression of Ret, Lhx1, and Sall4 in Glis3KO2 testis. Our study identifies Glis3 as a novel and critical regulator of early stages of spermatogenesis.

Thy1+ cells were isolated from 3 WT and 3 Glis3KO2 testis at postnatal day 4, and total RNAs were purified from them. Then the samples were applied to Agilent mouse genome chip.

Project description:Gene expression during spermatogenesis is highly variable and this differential pattern is very important for the successive culmination of different stages of the process, leading to production of the male gamete. Taking the time window of first wave of spermatogenesis, we did a microarray profiling of total testicular transcriptome in mouse and found several significant patterns of variable gene expression, forming upregulated and downregulated clusters among the three stages analyzed here. Mouse testis tissue were taken at three stages, 8 days, 16 days and 24 days, during first wave of spermatogenesis for RNA extraction and hybridization on the Affymetrix microarrays. Three biological replicates per age group were included and comparisons were done as 16 days vs 8 days, 24 days vs 16 days and 24 days vs 8 days testis transcriptome profile.

Project description:Vertebrate spermatogonial stem cells maintain sperm production over the lifetime of an animal but fertility declines with age. While morphological studies have informed our understanding of typical spermatogenesis, the molecular and cellular mechanisms underlying the maintenance and decline of spermatogenesis are not yet understood. We used single-cell RNA sequencing to generate a developmental atlas of the aging zebrafish testis. All testes contained spermatogonia, but we observed a progressive decline in spermatogenesis that correlates with age. Testes from some older males only contained spermatogonia and a reduced population of spermatocytes. Spermatogonia in older males are transcriptionally distinct from spermatogonia in testes capable of robust spermatogenesis. Immune cells including macrophages and lymphocytes drastically increase in abundance in testes that cannot complete spermatogenesis. Our developmental atlas reveals the cellular changes as the testis ages and defines a molecular roadmap for the regulation of spermatogenesis.

Project description:Gene expression in mammalian testis undergoing spermatogenesis is under strict post transcriptional regulation and microRNAs are known to play a role in this regulation. Considering the time window of first wave of spermatogenesis, we did a microarray profiling of total testicular microRNAs in mouse and found several significant patterns of variable expression of these molecules during the different stages analyzed here. Mouse testis tissue were taken at three stages, 8 days, 16 days and 24 days, during first wave of spermatogenesis for RNA extraction and hybridization on Locked Nucleic Acid(LNA) technology based Exiqon microarray platform. We included two biological replicates per age group and comparisons were done between the samples with respect to their hybridization to the 'reference' sample which had all the six samples pooled together.
