Extracellular vesicles proteomic profile in anti-TNF immunosuppressed mice: insights into protein dysregulation during Leishmania infantum infection

Project description:Visceral leishmaniasis (VL) is most frequent in immunosuppressed patients, especially among those receiving immunosuppressive therapy for autoimmune diseases, being TNF antagonist (anti-TNF) one of the main immunosuppressants used. Under these conditions, VL is most severe, with lower response to traditional leishmanicidal treatments such as antimonials (Sb) and a higher risk of relapse. Extracellular vesicles (EVs) are a great source of biological molecules, including proteins, capable of modulating immune-mediated cells and virulence factors. To identify proteins involved in the clinic of VL under anti-TNF immunosuppression, the proteomic profile of EVs from the serum of BALB/c mice was analysed using a label-free quantification (LFQ) proteomics approach. In infected mice with Leishmania infantum a comparison was made between immunosuppression with anti-TNF at clinical doses and non-immunosuppressed conditions, before and after treatment with Sb. Proteomics analysis revealed 271 dysregulated proteins in the pre-treatment groups, the majority of which were downregulated in the anti-TNF samples. On the other hand, 216 proteins were identified in the treated groups, being most of them enriched in the anti-TNF treated (anti-TNF+Sb) samples. The gene ontology (GO) analysis of these proteins revealed altered pathways related to the immune system, liver regeneration, or ion transport. Our results show dysregulated proteins associated with anti-TNF immunosuppression, which could have an impact on the greatest severity of the VL infection experienced in this situation. This study is an important step in the understanding of the clinical management of these VL pharmacologically immunosuppressed cases.

Differential protein secretome induced by TNF-alpha

Project description:Endothelial-mesenchymal-transition (EndMT) is an important source of cancer-associated fibroblasts (CAFs), which are known to facilitate tumor progression. We have previously shown that EndMT is present in pancreatic tumors and that deficiency of the Tie1 receptor induces EndMT in human endothelial cells. Pancreatic tumors are characterized by the presence of tumor necrosis factor-α (TNF-α). We now show that TNF-α strongly induces human endothelial cells to undergo EndMT. In order to know the secretory feature of cells which undergo EndMT by TNF-α, we conducted a comparative analysis of HMVEC secretome treated or not for 24h and 48h with TNF-α. Secretome study shows that cells treated with TNF-α have an important fibroblast-like secretory capacity, and a proinflamatory signature. Moreover, Ingenuity Pathway Analysis (IPA) shows that pathways implicated in migration, inflammation and fibrosis are predicted to be activated and that necrosis and apoptosis pathways are inhibited. Accordingly cell survival, viability and cycle progression are activated. We show that TNF-α- treated cells secrete proteins related to 16 protumoral pathways, confirming their fibroblastic characteristic. Finally, among the predicted upstream regulators activated, IPA analysis shows that, TNFSF12 and its receptor are present at hight levels in PDAC patients. Altogether these results show the fibroblastic characteristic of treated cells and demonstrate that TNF-α induces CAFs.

Expression data from TNF-stimulated mouse glomeruli

Project description:The specific contribution of the two TNF-receptors Tnfr1 and Tnfr2 to TNF-induced inflammation in the glomerulus is unknown. In mice, TNF exposure induces glomerular expression of inflammatory mediators like adhesion molecules and chemokines in vivo, and glomerular accumulation of leukocytes. To examine Tnfr-specific inflammatory responses in intrinsic glomerular cells but not infiltrating leukocytes we performed microarray gene expression profiling on intact glomeruli isolated from wild-type and Tnfr-deficient mice following TNF exposure in vitro. Glomeruli were isolated from male C57BL/6J wild-type and Tnfr-deficient mice applying a magnetic bead-based isolation technique. Isolated intact glomeruli were stimulated with TNF for 12 hours in vitro for subsequent RNA extraction and hybridization on Affymetrix microarrays.
