Project description:Background & Aims: Previous studies have suggested that dietary folic acid (FA) can protect against certain types of cancers. However, the findings have varied and the mechanisms by which this vitamin exerts chemopreventive effects remain to be clarified. We examined the effects of FA supplementation on DNA methylation, gene expression and gastric dysplasia in a transgenic mouse model that is etiologically and histologically well matched with human gastric cancers. Methods: Hypergastrinemic mice (INS-GAS) infected with Helicobacter felis were studied at multiple stages of gastric dysplasia and early cancer, with FA supplementation initiated both at weaning and later in life. Global DNA methylation was assessed by a methylation-sensitive cytosine incorporation assay, bisulfite pyrosequencing of B1 repetitive elements and immunohistochemistry (IHC) with anti-5-methylcytosine. We also profiled gene expression in the same tissues. Results: We found a decrease in global DNA methylation and tissue folate and an increase in serum homocysteine with progression of gastric dysplasia. FA supplementation prevented this loss of global DNA methylation and markedly reduced gastric dysplasia and mucosal inflammation. FA protected against the loss of global DNA methylation both in the dysplastic gastric epithelial cells and in gastric stromal myofibroblasts. In addition, FA supplementation had an anti-inflammatory effect, as indicated by expression profiling and IHC for lymphocyte markers. Conclusions: We conclude that FA supplementation is chemopreventive in this model of Helicobacter-associated gastric cancer. The beneficial effect of FA is likely due to its ability to reverse global loss of methylation and suppress inflammation. Study the difference in gene expression between transgenic hypergastrinemic (INS-GAS) mice fed a folate-supplemented diet and a diet with normal folate content. Because folate had a substantial protective effect, we matched the animals not only by folate status but also by dysplasia score (DYS). The degree of dysplasia was scored on a 1-4 scale. True replicates are not included. The first five samples are from folate supplemented animals, and the latter five are from controls on regular chow.

Project description:Many vaccination studies have revealed various degrees of protection in mouse models. However, the mechanism of protection is not fully understood. The aim of this study was to identify genes specifically expressed in H. pylori infection and prophylactic immunized mice. A prophylactic immunization with purified protein of H. pylori BabA combining with the mucosal CTA1-DD adjuvant was performed in a-1,3/4-fucosyltransferase transgenic FVB/N mice. Gene expression profile was analysed from the gastric mucosa among untreated, the infected and immunized mice. There were a set of genes were upregulated or downregulated respectively, in infected mice as compared with untreated mice. These include the adipokines released form adipose tissue, adhesion molecules and actin cytoskeletal rearragemet associated genes. In immunized mice, however, the host response was not as strong as in infected mice and significantly changed genes were completely overlapped with those in infected mice. The expression data from genechip was confirmed by quantitative real-time PCR assay. The microarray analysis suggests that genes such as actin cytoskeletal molecules and adipokines may be as potential targets of H. pylori infection. Prophylactic immunization with purified protein of H. pylori BabA combining with the mucosal CTA1-DD adjuvant could reduce bacteria colonization without induced a severe degree of inflammation. Project description:Focal and diffuse damage to the gastric mucosa contributes to gastric mucosal injury (GMI). Dietary fiber may exert protective effects against GMI; however, its use as a gastrointestinal intervention remains limited. Here, we assessed the potential role of dietary fiber Riclin in GMI. Dietary fiber Riclin exerted a dual effect in enhancing gastric mucosal defense. It modulated microbial-metabolite composition, enhancing biological barriers and reinforcing sustained adhesion to the gastric mucus bicarbonate barrier. Additionally, Riclin enhanced the gastric mucosal immune barrier through the NOD/TLR4/NF-κB pathway. The protective role of Riclin in GMI was revealed by constructing an alcohol-induced murine gastric ulcer model. Our results propose Riclin as a novel dietary fiber with the potential to ameliorate a wide range of gastric disorders characterized by a disrupted gastric mucosal defense and immune dysregulation. In particular, we underscore its potential as a promising dietary fiber for promoting gastric health.

Project description:BACKGROUND & AIMS: Gastric cancer is the second most frequent cause of death from cancer in the world, diffuse-type gastric cancer (DGC) exhibiting a poor prognosis. Germline mutations of CDH1, encoding E-cadherin, have been reported in hereditary DGCs, and genetic and/or epigenetic alterations of CDH1 are frequently detected in sporadic DGCs. Genetic alterations of TP53 are also frequently found in DGCs. To examine the synergistic effect of loss of E-cadherin and p53 on gastric carcinogenesis, we established a mouse line in which E-cadherin and p53 are specifically inactivated in the stomach parietal cell lineage. METHODS: We crossed Atp4b-Cre mice with Cdh1loxP/loxP and Trp53loxP/loxP mice, and examined the gastric phenotype of Atp4b-Cre+;Cdh1loxP/loxP;Trp53loxP/loxP mice. RESULTS: Non-polarization of E-cadherin-negative parietal cells and proton pump-negative atypical foci were observed in the transgenic mice. Intramucosal cancers and invasive cancers composed of poorly differentiated carcinoma cells and signet ring cells, which were histologically very similar to those in humans, were found from 6 and 9 months, respectively. Fatal DGCs developed at 100% penetrance within a year, frequently metastasized to lymph nodes, and had tumorigenic activity in immunodeficient mice. Gene expression profiling analyses also revealed that DGCs in the E-cadherin/p53-deficient mice resembled human DGCs. CONCLUSIONS: Our mouse line is the first genetically modified mouse model of DGC and very useful for clarifying the mechanism underlying gastric carcinogenesis, and provides a new approach to the treatment and prevention of DGC because of morphological and biochemical similarities with human DGC. Project description:YAP and TAZ play oncogenic roles in various organs, but the role of YAP/TAZ activation in gastric cancer in vivo has been understudied. We investigated whether and how YAP/TAZ initiates gastric tumorigenesis in vivo and its significance in human gastric cancer. We studied Lats1fl/fl;Lats2fl/fl;Lgr5-CreER mice, which have activated YAP/TAZ in pyloric stem cells. Gastric tissues were collected and analyzed by histopathology and immunostaining. To scrutinize the molecular mechanisms, we performed RNA sequencing in gastric tissue samples. Tissue microarray and public transcriptome data were analyzed to investigate the importance of YAP/TAZ in human gastric cancer.
