Proteome of oxytetracycline-resistant Edwardsiella tarda

ABSTRACT: We obtained a strain of oxytetracycline-resistant Edwardsiella tarda named LTB4-ROTC through artificial passaging in the TSB medium，the minimum inhibitory concentration (MIC) of LTB4-ROTC was 12.5 μg/mL, which was 32 folds increase contrast to LTB4-S strain. Proteomic analysis was conducted to profile the different protein expression.

Project description:we propagate norfloxacin-resistant strain, LTB4-R, through sequential propagations of norfloxacin-sensitive LTB4 in TSB broth medium with norfloxacin. LTB4-R had 128-fold minimum inhibitory concentrations (MIC) of LTB4-S. To profile the differential protein expression between LTB4-R and LTB4-S, tandem mass tag (TMT)-based quantitative proteomics method was adopted.

Acinetobacter baumannii response to cefiderocol challenge in human urine

Project description:Cefiderocol (CFDC) is a novel chlorocatechol-substituted siderophore approved to treat complicated urinary tract infections and for hospital-acquired and ventilator-acquired pneumonia. In previous work, human fluids, were shown to increase the minimum inhibitory concentration (MICs) of Acinetobacter baumannii against CFDC and reduce the expression of genes related to iron uptake systems, which could explain the need for higher concentrations of CFDC to exert inhibitory action. Herein, we analyzed the impact of human urine (HU), which contains low albumin concentrations, on the expression of iron-uptake related genes and MIC values of two carbapenem-resistant A. baumannii. Levels of resistance to CFDC were not modified by HU in strain AMA40 but were reduced in the case of strain AB5075. Testing other carbapenem-resistant A. baumannii isolates showed that the CFDC MICs were unmodified or reduced in the presence of HU. The expression of piuA, pirA, bauA, and bfnH determined by qRT-PCR was enhanced in both strains when HU was present in the culture medium. All four tested genes are involved in recognizing ferric siderophore complexes or internalization into the cell’s cytosol. In contrast, the effect of HU on genes associated with resistance to β-lactams, antibiotics commonly used to treat urinary tract infections caused by A. baumannii, was variable; the transcriptional analysis of pbp1, pbp3, blaOXA-51-like, blaADC, and blaNDM-1 showed significant variation. In summary, HU, probably due to the albumin and free iron content, does not adversely impact or slightly improves the activity of CFDC when tested against A. baumannii in urine in contrast to other human bodily fluids.

The transcriptional response of Pasteurella multocida to three classes of antibiotics

Project description:We studied the effects of three classes of antibiotics (amoxicillin, chlortetracycline and enrofloxacin ) on P. multocida transcriptome using custom oligonucleotide microarrays from Nimblegen systems. All the 2015 genes of Pm70 were spotted on the array and hybridizations were carried out with RNA isolated from three independent cultures of Pm70 grown in the presence or absence of sub-minimum inhibitory (sub-MIC) doses of antibiotics. Differentially expressed genes were identified by ANOVA and Dunnett’s test. Biological modeling of the differentially expressed genes (DE) was carried out based on Clusters of Orthologous (COG) groups and network analysis in Pathway Studio. Keywords: Response to sub-MIC antibiotics The experimental design included three biological replicate cultures of P. multocida grown in the absence or presence of sub-MIC antibiotics. Effects of antibiotics on the transcriptome with each antibiotic were determined by comparing the growth in the presence of antibiotic (treatment) to growth in the absence of antibiotic (control).

Aspartate potentiates tobramycin against multidrug-resistant Edwardsiella tarda through enhancing proton motive force and membrane permeability

Project description:<p>The antibiotic susceptibility/resistance of bacteria is influenced by their metabolic states. In tobramycin-resistant Edwardsiella tarda (LTB4-RToB), a dynamic metabolic state is observed, characterized by a general reduction in most metabolites as the minimum inhibitory concentration (MIC) increases. Among these, aspartate downregulation emerges as the most critical biomarker. Exogenous aspartate enhances the efficacy of tobramycin, enabling it to effectively kill both lab-evolved and clinically isolated multidrug-resistant (MDR) E. tarda, as demonstrated in an animal model. Aspartate increases intracellular tobramycin levels in a concentration- and time-dependent manner by promoting drug uptake. Metabolic reprogramming analysis of LTB4-RToB and clinically isolated MDR strain WY-28 reveals that exogenous aspartate restores downregulated metabolites, primarily enhancing alanine, aspartate, and glutamate metabolism; the pyruvate cycle; and glycine, serine, and threonine metabolism. These changes elevate the proton motive force and membrane permeability, thereby increasing tobramycin uptake. The resulting intracellular drug concentration surpasses the bactericidal threshold, effectively overcoming resistance in both LTB4-RroB and clinical MDR E. tarda. These findings identify aspartate as a potent metabolic reprogramming agent that potentiates aminoglycosides by boosting intracellular drug accumulation, offering a strategy to combat antibiotic-resistant E. tarda.</p>

Analysis of antibiotic response in Clinical Wound Pseudomonas aeruginosa isolates: Unveiling Proteome Dynamics of tobramycin tolerant phenotype

Project description:Pseudomonas aeruginosa (PA) is an opportunistic human pathogen, causing serious chronic infections. PA can adapt efficiently to antibiotic stressors via different genotypic or phenotypic strategies such as resistance and tolerance. The adaptation regulatory system is not always very well understood. In this study, we use shotgun proteomics to investigate the system-level response to tobramycin in two clinical wound PA isolates and PAO1. We profiled each strain for its antibiotic drug-tolerant phenotype using supra-minimum inhibitory concentrations (supra-MIC) of tobramycin and apply proteomics to investigate the protein expression profiles. The MIC revealed that all isolates were susceptible to tobramycin but at supra-MIC concentrations at stationary growth, a degree of tolerance was observed for the isolates. We identified around 40 % of the total proteins encoded by the PA genome and highlighted shared and unique protein signatures for all isolates. Comparative proteome profiling in the absence of antibiotic treatment showed divergent fingerprints, despite similarities in the growth behavior of the isolates. In the presence of tobramycin, the isolates shared a common response in the downregulation of proteins involved in the two-component system, whereas stress response proteins were present at higher levels. Our findings provide insight into the use of proteomic tools to dissect the system-level response in clinical isolates in the absence and presence of antibiotic stress.
