ABSTRACT: The interactome of FVIII-BDD-WT and FVIII-BDD-RH in HepG.A total of 1587 proteins were reproducibly detected among the biological replicates and of these, 177 proteins were differentially expressed with a p value of less than 0.05 and a fold change value of greater than 1.5.2 cells by IP-MS analysis.

Inosine Induces Stemness Features in HA-CAR-T cells and Enhances Potency [RNA-seq]

Project description:Adenosine (Ado) mediates immune suppression in the tumor microenvironment and exhausted CD8+ HA-CAR-T cells commonly express CD39 and CD73, which mediate proximal steps in Ado generation. Here we sought to enhance HA-CAR-T cell potency by knocking out CD39, CD73 or adenosine receptor 2a (A2aR), but observed only modest effects. In contrast, overexpression of Ado deaminase (ADA-OE), which metabolizes Ado to inosine (INO), induced stemness features and potently enhanced HA-CAR-T functionality. Similarly, and to a greater extent, exposure of HA-CAR-T cells to INO augmented HA-CAR-T cell function and induced hallmark features of T cell stemness. INO induced profound metabolic reprogramming, diminishing glycolysis and increasing oxidative phosphorylation, glutaminolysis and polyamine synthesis, and reprogrammed the epigenome toward greater stemness. Clinical scale manufacturing using INO generated enhanced potency HA-CAR-T cell products meeting criteria for clinical dosing. These results identify INO as a potent modulator of T cell metabolism and epigenetic stemness programming and deliver a new enhanced potency platform for immune cell manufacturing.

Global gene expression differences between GT1, RH-ERP2009 and RH-JSR

Project description:Previous studies have shown that continuous in vitro passage can result in changes to the global gene expression profiles in mammalian cells. To examine the extent of gene expression differences between highly homologous type I strains, we took advantage of the T. gondii Affymetrix microarray containing probes to more than 8,000 genes. We compared the RH-JSR and GT1 strains, two isolates that share the small plaque phenotype, to the large plaque isolate RH-ERP2009. Following hybridization and normalization of data, we used two independent methods to generate lists of genes with expression differences that were statistically significant and 2-fold or greater in at least one sample. The 1-way ANOVA (P ≤ 0.05) identified 520 genes that were significantly differentially expressed between GT1, RH-ERP2009 and RH-JSR. Statistical Analysis of Microarray (SAM) (FDR = .05%) analysis identified 475 transcripts that were differentially expressed. Both lists were combined (610 genes) and used to perform pairwise fold comparisons between the three samples. This process identified 113 genes that were similarly expressed in GT1 and RH-JSR and differentially expressed in RH-ERP2009. Differentially expressed genes were located across all chromosomes and showed no pattern of clustering to one particular region of the genome. Interestingly, three out of 34 ABC transporters (ToxoDB) were significantly upregulated in RH-ERP2009 (hypergeometric distribution, P = 0.01)

Chlorella vulgaris Protein Isolate Effectively Protects Lacticaseibacillus rhamnosus GG Viability During processing, Storage, and In vitro Digestion

Project description:This study examined the protective effect of Chlorella vulgaris protein isolate (CPI) on the biological activity of Lacticaseibacillus rhamnosus GG (LGG) during lyophilisation, storage, and in vitro digestion. Prior to lyophilisation, probiotic hydrogels were either fermented to pH 4.5 (CF) or left untreated (CNT). The microstructure, physicochemical, and thermal properties of the resulting probiotic powders were analysed, along with LGG adhesion to an in vitro intestinal epithelium model. Embedment in CPI-based matrices provided effective lyoprotection, with CNT offering greater stability than CF. Elevated storage temperatures and relative humidity (RH) accelerated LGG inactivation, particularly in CF. While a sufficient proportion of LGG survived the harsh conditions during in vitro digestion, pre-fermentation had a negligible impact. Additionally, a satisfactory level of LGG adhesion to the intestinal mucus layer was observed. Overall, CPI exhibits strong stabilizing properties for LGG viability and represents a promising plant-based alternative to dairy proteins for probiotic food formulations.

Project description:We designed a highly emissive rhodium(III) complex ( Rh(III) complex) as the mtDNA intercalator. Rh(III) complex can specifically bind to mtDNA to cause the cytoplasmic release of mtDNA fragments to activate the cGAS-STING pathway. Moreover, Rh(III) complex activates the mitochondrial retrograde signaling by disturbing the key metabolites. Herein,we performed RNA-seq to illustrate the function of Rh(III) complex on HeLa cells. RNA-seq was performed on HeLa cells, which were treated by 1% DMSO or Rh(III) complex (1 uM, 6h), respectively.
