Project description:Transcriptome analysis in cotton under drought stress. To study the molecular response of drought stress in cotton under field condition global gene expression analysis was carried out in leaf tissue. Gossypium hirsutum cv. Bikaneri Nerma was used for the gene expression analysis. Cotton plants were subjected to drought stress at peak flowering stage. Leaf samples were collected when the soil moisture content was 19.5% which is 50% of the normal control plots. Gene expression profiles in drought induced and their respective control samples were analyzed using Affymertix cotton Genechip Genome arrays to study the global changes in the expression of genome.

Project description:Global gene expression was compared between roots of cotton plants (variety Sicot 71) flooded for 4 hours and roots of unflooded cotton plants. Global gene expression was also compared between leaves of cotton plants (variety Sicot 71) flooded for 24 hours and leaves of unflooded cotton plants. Waterlogging stress causes yield reductions in cotton (Gossypium hirsutum L.). A major component of waterlogging stress is the lack of oxygen available to submerged tissues. While changes in expressed protein, gene transcription and metabolite levels have been studied in response to low oxygen stress, little research has been done on molecular responses to waterlogging in cotton. We assessed cotton growth responses to waterlogging and assayed global gene transcription responses in root and leaf cotton tissues of partially submerged plants. Waterlogging causes significant reductions in stem elongation, shoot mass, root mass, and leaf number. At the global gene expression level waterlogging significantly alters the expression of 1012 genes (4.2% of genes assayed) in root tissue as early as 4h after flooding. Many of these genes are associated with cell wall modification and growth pathways, glycolysis, fermentation, mitochondrial electron transport and nitrogen metabolism. Waterlogging of plant roots also altered global leaf gene expression, significantly changing the expression of 1305 genes (5.4% of genes assayed) after 24h of flooding. Genes associated with cell wall growth and modification, tetrapyrrole synthesis, hormone response, starch metabolism and nitrogen metabolism were affected in leaf tissues of waterlogged plants. Implications of these results for the development of waterlogging tolerant cotton are discussed. Keywords: Stress Response

Project description:Cotton premature leaf senescence often occurred with an increasing frequency in many cotton growing areas and caused serious reduction in yield and quality of cotton has been one of the impontant factors that restrict severely the production of cotton.Our laboratory studies showed chilling stress is the key factor that induced A. alternatia infection, caused Alternaria disease and then lead to cotton leaf senescence, but the molecular mechanism of cotton premature leaf senscence is still unclear. We used microarrays to study molecular mechanism of chilling stress causing Alternaria alternata infection and leading to cotton leaf senescence and find the key genes during this process.

Project description:To comprehend the gene expression profile in rice flag leaf under high temperature, Agilent 4×44k rice oligo microarray experiments were carried out using rice flag leaf of reproductive stage at 0 min, 20 min, 60 min, 2 hr, 4 hr, and 8 hr after the treatment of 40 degree centigrade, and the significantly expressed genes mainly involved in transcriptional regulation, transport, protein binding, antioxidant, and stress response were identified. Among them, the predominant transcription factor gene families were Hsf, NAC, AP2/ERF, WRKY, MYB, and C2H2. KMC analysis discovered the time-dependent gene expression pattern under heat. The metabolism pathway analysis demonstrated that, under heat treatment, glycolysis and ubiquitin-proteasome was enhanced, and TCA, gluconeogenesis, the secondary metabolism and light-reaction in the photosystem was dramatically repressed, which revealed the great importance of maintaining primary metabolism and protein homeostasis in response to heat in rice flag leaf.

While, the normal growth plants were sustained growing at optimal temperature of 28/20â day/night. And then, both chilling stress pre-treated and normal growth cotton plants were inoculated with Alternaria. alternata isolate A1. The mock inoculations were performed with sterilized water. Cotton leaf Samples were respectively collected at 3, 6 days after inoculation (DAI) for RNA extraction and hybridization on Affymetrix microarrays. To that end, we collected 8 samples, i.e. chilling stress pre-treated cotton leaves: 3 DAI (C) and its mock control (D), 6 DAI (E) and its mock control (F); normal growth cotton leaves: 3 DAI (H) and its mock control (I), 6 DAI (J) and its mock control (K). All samples were arranged in completely randomized designed with three replications for each treatment.

Project description:Cotton (Gossypium hirsutum L.), a crucial global fiber and oil seed crop, faces a diverse biotic and abiotic stresses. Among these, temperature stress strongly influence its growth, prompting adaptive physiological, biochemical and molecular changes. In this study, we explored the proteomic changes underscoring the heat stress tolerance in the leaves of two locally developed cotton genotypes, i.e., heat tolerant (GH-Hamaliya Htol) and heat susceptible (CIM-789 Hsus), guided by morpho-physiological and biochemical analysis. These genotypes were sown at two different temperatures, control (35ºC) and stress (45ºC) in glass house, in randomized complete block design (RCBD) in three replications. At the flowering stage, a label-free quantitative shotgun proteomics of cotton leaves revealed the differential expression of 701 and 1270 proteins in the tolerant and susceptible genotype compared to the control, respectively. Physiological and biochemical analysis showed that the heat-tolerant genotype responded uniquely to stress by maintaining the net photosynthetic rate (Pn)( 25.2-17.5µmolCO2m-2S-1), chlorophyll (8.5-7.8mg/g FW), and proline contents (4.9-7.4 µmole/g) compared to control, supported by the upregulation of many proteins involved in several pathways including photosynthesis, oxidoreductase activity, response to stresses, translation, transporter activities as well asprotein and carbohydrate metabolic processes. In contrast, the distinctive pattern of protein downregulation involved in stress response, oxidoreductase activity, and carbohydrate metabolism was observed in susceptible plants. Thus, in this study, the specific proteins that increased in abundance under heat stress tolerance mechanism can be used as markers in future for producing the heat tolerant cotton genotypes without compromising the yield.

Project description:Five allotetraploid cotton species have adapted, through their transcriptional responses, to unique environments with distinct levels of inherent abiotic stresses. The transcriptional responses of leaf and root tissue in five allotetraploid cotton species (Gossypium hirsutum, G. barbadense, G. tomentosum, G. mustelinum, and G. darwinii) under salt stress have been investigated in this study using cotton long oligonucleotide microarrays. Physiological responses to salinity such as stomatal conductance, ion and osmoprotectant contents were also measured as indicators of imposed stress. Accessions from these five cotton species were hydroponically grown and gradually introduced to a NaCl treatment (15 dS m-1). The microarray results identified 2721 and 2460 differentially expressed genes under salt stress that were significant in leaf and root tissue, respectively. Many of these genes were classified under gene ontology (GO) categories that suggest abiotic stress. These allotetraploid cottons shared transcriptional responses to salinity, but also showed responses that were species-specific. No consistent differences in transcriptional response among the previously estimated phylogenetic branches were found. Stomatal conductance, ion accumulation, and betaine, trigonelline, and trehalose contents also indicated salt stress. This global assessment of transcriptional and physiological responses to salt stress of these cotton species may identify possible gene targets for crop improvement and evolutionary studies of cotton. Project description:Wheat (Triticum ssp.) is one of the most important human food sources as well as livestock feed, but is very sensitive to changes in temperature. Specifically, processes during leaf growth and photosynthesis and during flower and seed development are affected by high temperature. While this has been investigated to some extent on physiological, developmental and molecular levels, very little is known about early signalling associated with an increase in temperature. Here, we probed the impact of an increase in temperature for 1 hour on the leaf and flower phosphoproteome of wheat. In order to identify differentially phosphorylated peptides, we used two wheat proteome databases, and could show the superiority of the new IWGSC genome. Our analyses of the leaf phosphoproteome further revealed differential phosphorylation of heat shock proteins, Photosystem I subunits, and plasma membrane intrinsic proteins, in addition to a number of potentially novel players.
