The abnormal band for LRRC8A in SDA-PAGE gel was identified by MS

ABSTRACT: LRRC8A protein in the OE-LRRC8A group was overexpressed by more than 2.5-fold compared with that noted in the control and LV-ctrl groups (P < 0.05); three clear bands were identified in the OE-LRRC8A sample by western blotting, and the uppermost band was not shown in the specification of LRRC8A antibody. Then the product was extracted from the SDS-PAGE gel of the uppermost band location and analyzed by mass spectroscopy to confirm whether this band contains LRRC8A; the specific peptides of LRRC8A were captured and detected successfully. This indicated that the uppermost band contains LRRC8A protein.

The protein phosphorylation in HCT116 cells with or without the over-expressed LRRC8A

Project description:It’s hypothesized that LRRC8A may regulate cell proliferation and migration by participating in the transduction of cell signaling. Then protein phosphorylation in cells with or without the over-expressed LRRC8A was detected and quantified by mass spectroscopy. A total of 564 proteins and 1,102 phosphorylated sites were identified, of which 342 proteins and 538 phosphorylated sites were successfully quantified. A total of 32 downregulated and 68 upregulated proteins were noted in the OE-LRRC8A group compared with the LV-ctrl group; the intensities of 79 phosphorylated sites in 62 proteins were considerably altered by the overexpression of the LRRC8A protein; all the differential sites included 41 downregulated sites and 38 upregulated sites.

Load-Dependent Changes in Left Ventricular Gene Expression in a Pathophysiologically Relevant Murine Model of Reversible Heart Failure (2 weeks heart failure, 4 weeks reversal)

Project description:Wild type, female, C57BL/6 mice were subjected to sham (n=6) surgery, or TAC + MI to cause progressive LV remodeling (n=12). At 2wks post-TAC, one group of mice underwent de-banding (HF-DB, n=6), whereas in a second group of mice the band remained intact (HF; n = 6). LV remodeling was evaluated by 2D echocardiography at 14 days post-TAC+MI , and 4 wks post-debanding. At 6 wks the hearts were excised and analyzed for changes in gene expression using transcriptional profiling. e-banding in the HF-DB mice resulted in normalization of LV volumes and LV mass, and normalization of cardiac myocyte hypertrophy at 6wks, without significant changes in myofibrillar collagen in the HF and HF-DB mice. Both LV ejection fraction (LVEF) and LV radial strain improved numerically following de-banding; however, both measurements remained significantly depressed in the HF-DB mice compared to sham, and were not significantly different from HF mice at 6wks. Reverse LV remodeling in the HF-DB mouse hearts was accompanied by a partial (80%) normalization of the genes that became dysregulated during LV remodeling, whereas 20% of genes remained persistently dysregulated following reverse LV remodeling.

hsa-miRNA-548v controls the viscoelastic properties of human cardiomyocytes and improves their relaxation rates

Project description:The impairment of left ventricular (LV) diastolic function with inadequate increase in myocardial relaxation velocity directly results in lower LV compliance, increased LV filling pressures and heart failure symptoms. The development of agents facilitating the relaxation of human cardiomyocytes requires a better understanding of the underlying regulatory mechanisms. We performed a high-content microscopy-based screening in human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) using a library of 2565 human miRNA mimics and measured relaxation kinetics via high-computing analyses of motion movies. We identified hsa-miR-548v, a primate specific miRNA, as the miRNA producing the largest increase in relaxation velocities. This positive lusitropic effect was reproduced in engineered cardiac tissues generated with healthy and BRAF T599R mutant hiPSC-CMs, and was independent of changes in calcium transients. Consistent with improvements in viscoelastic responses to mechanical stretch, RNA-sequencing showed that hsa-miR-548v down-regulated multiple targets, especially components of the mechano-sensing machinery. The exogenous administration of hsa-miR-548v in hiPSC-CMs notably resulted in a significant reduction of ANKRD1/CARP1 expression and localization at the sarcomeric I-band. This study suggests that the sarcomere I-band is a critical control center of the ability of cardiomyocytes to relax and a target for improving relaxation and diastolic dysfunction.
