Analysis of mRNA expression in human skin fibroblasts WS1 cells between IFI6-overexpression and control upon irradiation.

Project description:This study aims to compared mRNA expression between IFI6-overexpressed WS1 cells and NC by RNA-Seq. In this study, WS1 cells were divided into two groups and each group had four repeats, one group was infected by IFI6-overexpressed lentivirus and another was infected by NC lentivirus. The two groups of cells were then irradiated with a single 5 Gy dose of radiation at a fixed rate of 1.15 Gy/min using an X-ray linear accelerator (Rad Source Technologies Inc., Suwanee, GA, USA). 24 hours post irradiation, WS1 cells from IFI6-overexpression and NC were collected and subjected to mRNA expression analysis.

Transcriptome profiling of de-differentiated chondrocytes over-expressing miR-138 or miR-181ab1 during osteogenic induction

Project description:The goal of this study was to investigate the effects of miR-138 or miR-181ab1 on regulating osteogenesis. Gene expression profiles were analyzed in human de-differentiated chondrocytes (DDCs) over-expressing either miR-138 or miR-181ab1 following day 2 or day 7 of osteogenic induction. DDCs were isolated from human osteoarthritic articular cartilage and over-expression of miR-138 or miR-181ab1 was induced following lentiviral transduction of DDCs over-expressing the precursor form of miR-138 or miR-181ab1. Control DDCs were transduced with lentivirus expressing a non-silencing (NS) control RNA. Total RNA was harvested at day 2 or day 7 following osteogenic induction and subjected to RNA-Seq analysis.

Novel targets of microRNA miR-142 in cardiac myocytes

Project description:The nuclear acetyltransferase p300 is a dose-dependent and limiting regulator of cardiac hypertrophy. p300 regulates hypertrophy by controlling the expression of specific microRNAs.The objective of this study was to dissect the role of miR-142 during p300 regulated hypertophic growth in vitro and in vivo and to verify its targets. MiR142 and a non target control were overexpressed using lentivirus in primary culture of neonatal cardiac myocytes. Overexpression was checked 48 hour post transfection. Samples include miR142 OE and non target from three different experiments.
