Proteomic analysis of PDCoV-infected HIEC-6 cells


ABSTRACT: Data-independent acquisition (DIA) is a new type of mass spectrometry data acquisition developed in recent years. The entire scanning range of mass spectrometry can be divided into several windows based on the mass-to-charge ratio (m/z), and then all the parent ions in each window are fragmented and detected, and the fragment ion information of all the parent ions is collected for protein characterization and quantification. Currently popular proteomics research tools, such as iTRAQ/TMT, Label-free, SILAC use a data-dependent acquisition (DDA) mode of scanning, the first level of mass spectrometry for full scanning acquisition of parent ion signals to generate the first level of spectrum, the second level of mass spectrometry to select the first level of the signal intensity in the spectrum in turn In the secondary mass spectrometry, the peaks ranked as top10/20/40 in the primary spectrum were fragmented and the corresponding daughter ions were collected.
