[HPP-2.0] Indentification of biomarkers and therapeutic targets for esophageal cancer

ABSTRACT: We established a cohort of 180 patients undergoing surgery for esophageal cancer and analyzed the samples using proteomic techniques to indentify biomarkers and therapeutic targets for esophageal cancer and further explore how these factors modulate the immune microenvironment and mediate the specific mechanism of immunotherapy response or drug resistance.

Project description:We established a cohort of neoadjuvant immunotherapy for esophageal cancer, using proteomics to obtain protein molecules associated with efficacy and capable of characterizing the microenvironment of esophageal cancer and verify their effect in predicting immunotherapy response in an independent neoadjuvant immunotherapy cohort or in the real world. Future prospective clinical trials will be conducted to verify the effect of screening populations.

MicroRNAs among esophageal cancer cell sublines with different migration ability

Project description:Cellular directed migration is critical to invasion-metastasis cascade of cancer cells. We used in vitro transwell model to screen two esophageal cancer cell lines (KYSE30/180) and obtained two pairs of subpopulations with distinc motiliyt ability. Then we used microarrays to detail the differentially expressed genes and microRNAs between these two cell sublines (30U/D and 180U/D) to identify those responsible for ESCC motility. KYSE30/180 cells were subject to four successive selections using transwell (CORNING, USA). Subpopulations penetrated through membrane (D) or not (U) were harvested respectively for RNA extraction and hybridization on Affymetrix genome and LC Sciences microRNA microarrays

Transcription profiling of human HDAC-42 SEG-1 esophageal adenocarcinoma cells

Project description:The role of acid and bile salts in the pathogenesis of esophageal carcinoma arising from Barrett's metaplasia has been well established. Cell proliferation of Barrett's epithelium in response to pulsatile acid exposure has since been confirmed in vivo using endoscopy specimens. Histone deactylases (HDACs) modulate nucleosomal packaging of DNA, thereby influencing gene transcription and multiple cancer-associated processes. Thus, we conducted microarray analysis to assess the ability of HDAC-42 to modulate key acid-induced changes as well as to impact other genes altered as the normal esophageal epithelium progresses along the metaplasia-dysplasia-esophageal adenocarcinoma continuum. Experiment Overall Design: SEG-1 cells were pretreated for 24 hours with vehicle or HDAC-42, pulsed with media or acidified media (pH 3.5; 20 mins at 37C), replenished with media or HDAC-42, and harvested 6 hours later. Global gene expression analysis was conducted using the human genome chip U133 2.0 Plus.

Comparative single-cell analysis of esophageal cancer subtypes reveals cellular distinctions explaining varied immunotherapy responses

Project description:Esophageal cancer comprises two main subtypes: esophageal squamous cell carcinoma (ESCC) and esophageal adenocarcinoma (EAC). Previous studies revealed their distinct genomic characteristics, sharing similarities with head and neck squamous cell carcinoma (HNSCC) and gastric adenocarcinoma (GAC) for ESCC and EAC, respectively. This study compares ESCC and EAC at single-cell resolution to identify factors explaining differences in immunotherapy response rates. We performed comparative single-cell transcriptome analysis for four cancer types near the esophagus using data from 35 patients with ESCC, EAC, HNSCC, or GAC. Malignant epithelial cells showed distinct separations between subtypes based on histological origins. We identified enrichment of CXCL13+ CD8+ T cells and CXCL9+CXCL10+ TAMs in ESCC and HNSCC, promoting a hot-tumor environment through interferon-γ pathways. Conversely, hypoxia and cold-tumor related populations, such as heat-shock protein high CD8+ T cells and MARCO+ TAMs, were enriched in EAC and GAC. These immune subsets' expression signatures predicted immunotherapy responses across diverse cancer types.
