ABSTRACT: Stenotrophomonas sp. SY1 showed the highly efficient detoxification to cadmium and chromium. Thus, the purpose of this project is to clarify the mechanism of detoxification to cadmium and chromium in strain SY1.

Genome-wide transcriptional responses of two metal-tolerant symbiotic Mesorhizobium isolates to zinc and cadmium exposure

Project description:Mesorhizobium metallidurans STM 2683 and Mesorhizobium sp. strain STM 4661 were isolated from nodules of the metallicolous legume Anthyllis vulneraria from distant mining spoils. They tolerate unusually high zinc and cadmium concentrations as compared to other mesorhizobia. This work aims to study the gene expression profiles associated with zinc or cadmium exposure and to identify genes involved in metal tolerance in these two metallicolous Mesorhizobium strains of interest for mine phytostabilization purposes. Mesorhizobium metallidurans STM 2683 and Mesorhizobium sp. strain STM 4661 with three treatments (control, Zn and Cd).

Drivers of metallothionein-mediated detoxification of cadmium in C.elegans

Project description:Cadmium is toxic and intricate pathways linked to metallothioneins (MT) drive the detoxification process. Whist the mechanisms are well understood in mammalian systems, detailed knowledge is still elusive in invertebrates (which notably differ to mammalian systems). The model nematode Caenorhabditis elegans is ideally suited for assessing metallothionein mediates detoxification in invertebrates as is relatively short-lived, can be easily exposed and its genome is fully sequenced and widely annotated. The aim of the experiment was to identify new MT mediated target genes involved in Cd toxicosis. The global transcriptome was compared in wild type and a MT double knockout strain raised in the presence or absence of 30 µM Cd.

Threonine dehydratase couples L-cysteine metabolism to cadmium resistance in Pseudomonas stutzeri 273 through cadmium sulfide nanoparticle biomineralization

Project description:Cysteine is an amino acid containing sulfhydryl (-SH) group which is the basis of cysteine involved in sulfur metabolism and heavy metal detoxification in microorganisms. Here, we demonstrate that adding L-cysteine significantly improves the cadmium resistance and removal ability of Pseudomonas stutzeri 273. Threonine dehydratase (TSD) connects L-cysteine metabolism and cadmium resistance through CdS nanoparticle biomineralization with the ability of catalyzing L-cysteine desulfuration and H2S generation in P. stutzeri 273. Gene knockout of TSD in P. stutzeri 273 results in the decrease of L-cysteine and cadmium resistance, decline of H2S generation, and reduction of CdS biosynthesis ability.
