Project description:In the current study, we compared phosphoproteomic and proteomic changes in roots of different soybean seedlings of a salt-tolerant cultivar (Wenfeng07) and a salt-sensitive cultivar (Union85140) induced by salt stress. The root samples of Wenfeng07 and Union85140 at three-trifoliate stage were collected at 0 hr, 0.5 hr, 1 hr, 4 hrs, 12 hrs, 24 hrs and 48 hrs after been treated with 150 mM NaCl. LC-MS/MS based phosphoproteomic analysis of these samples identified a total of 2692 phosphoproteins and 5509 phosphorylation sites. Of these, 2344 phosphoproteins containing 3744 phosphorylation sites were quantitatively analyzed. Our results showed that 1163 phosphorylation sites were differentially phosphorylated in the two compared cultivars.

Project description:To identify potential substrates of STK and their target phosphorylation sites in SS2, phosphoproteomic analysis was performed. A comparison of phosphorylated peptides from the WT strain (ZY05719) to those in the mutant strain (Δstk) helped us to identify phosphopeptides that disappeared or decreased in the mutant strain Δstk and are likely substrates of STK. In total, 41 unique phosphopeptides from 31 proteins were identified in S. suis lysates. Among the identified phosphopeptides, 44 phosphorylation sites were identified: 16 on serine, 23 on threonine and 5 on tyrosine. The phosphopeptides were considered to be phosphorylated by STK. Eleven phosphopeptides corresponding to 9 proteins had decreased phosphorylation in the Δstk strain. Twenty phosphopeptides corresponding to 14 proteins were only identified in the wild-type strain ZY05719. Because of the level of phosphorylation changes, these proteins may be STK target proteins that are phosphorylated in vivo.

Project description:Photosynthesis is affected by water deficiency (WD) stress, and nitric oxide (NO) is a free radical that participates in the photosynthesis process. Previous studies have suggested that NO regulates the excitation energy distribution of photosynthesis under WD stress. Here, quantitative phosphoproteomic profiling was conducted using isobaric tags for relative and absolute quantitation. Differentially phosphorylated protein species (DEPs) were identified in leaves of NO or polyethylene glycol (PEG)-treated wheat seedlings (D) and in control seedlings, 2,257 unique phosphorylated peptides and 2,416 phosphorylation sites were identified from 1,396 unique phosphoproteins. Of these, 96 DEPs displayed significant changes (≥ 1.50-fold, p < 0.01). These DEPs are involved in photosynthesis and signal transduction, etc. Furthermore, phosphorylation of several DEPs were up-regulated by both D and NO treatments, but down-regulated only in NO treatment. These differences affected the chlorophyll A-B binding protein, chloroplast post-illumination chlorophyll fluorescence increase protein, and SNT7, implying that NO indirectly regulated the absorption and transport of light energy in photosynthesis in response to WD stress. The significant difference of chlorophyll (Chl) content, Chl a fluorescence transient, photosynthesis index, and trapping and transport of light energy further indicated that exogenous NO under D stress enhanced the primary reaction of photosynthesis compared to D treatment. A putative pathway is proposed to elucidate NO regulation of the primary reaction of photosynthesis under WD.

Project description:This study aims to analyze and compare the post-translational modification differences between human plasma- and genetic recombination-derived human serum albumin (HSA) in clinical treatments in China. 6 different post-translational modifications, including acetylation, succinylation, crotonylation, phosphorylation, beta-hydroxybutyrylation, and lactylation, were detected using pan-specific antibodies through Western blot. The samples, consisting of human plasma-derived HSA (pHSA) from six different manufacturers and recombinant HSA (rHSA) expressed in yeast and oryza sativa, underwent detection of types of post-translational modifications. 4D label-free PTMs quantitative proteomic analysis was conducted to detect N-glycosylation and the above-mentioned post-translational modifications in pHSA and rHSA samples, and analyze the differences in modification sites and levels. Through Western blot analysis, all six pHSA and two rHSA samples were positive in the band of albumin (66.5 kDa) for the six post-translational modifications. Further analysis using 4D label-free PTMs quantitative proteomics revealed 25 (29) acetylated, 30 (32) succinylated, 41 (50) malonylated, 15 (23) phosphorylated, 36 (30) beta-hydroxybutyrylated, and 27 (34) lactylated modification sites in pHSA and rHSA samples and no N-glycosylation modification sites. The analysis results identified 1 acetylation (ALB_K160), 2 beta-hydroxybutyrylation (ALB_K569, ALB_K426), 3 crotonylation (ALB_K264, ALB_K581, ALB_K560), and 15 phosphorylation specific modification sites in pHSA, as well as 3 crotonylation (ALB_K560, ALB_K562, ALB_K75), 1 succinylation (ALB_K490), and 23 phosphorylation specific modification sites in rHSA. In pHSA (rHSA), 2 (6) acetylation, 10 (12) succinylation, 0 (9) crotonylation, 1 (9) phosphorylation, 6 (0) beta-hydroxybutyrylation, and 0 (7) lactylation specific modification sites were found. Additionally, in the common modification sites of pHSA and rHSA, pHSA showed up-regulation of amberylation (16:1) and beta-hydroxybutyrylation (12:2) in more sites, and up-regulation of acetylation (7:11), crotonylation (2:11), phosphorylation (1:8), and lactylation (1:14) in less sites compared to rHSA. In clinical treatment, the used pHSA and rHSA in China generally exhibit acetylation, succinylation, crotonylation, phosphorylation, beta-hydroxybutyrylation, and lactylation, and there are differences in the site characteristics and modification levels of these modifications between pHSA and rHSA. Further experimental investigations are needed to explore the impact of post-translational modification differences on the biological function, efficacy, and safety of pHSA and rHSA.

Project description:Purpose: The purpose of present study is to characterize the Idiopathic epiretinal membrane (iERM) through phosphoproteomics to facilitate its diagnosis and treatment. Experimental design: The vitreous of 25 patients with iERM and 15 patients with idiopathic macular hole were analyzed by proteomic and phosphoproteomic analysis based on tandem mass tag. PRM was used to verify differential proteins. Results: Proteomic analysis identified a total of 878 proteins, including 50 differential proteins. Phosphoproteomic analysis identified a total of 401 phosphorylation sites on 213 proteins, including 27 differential phosphorylation sites on 24 proteins. Twenty six proteins of these differential proteins and phosphorylated proteins may be closely related to fibrosis in iERM. MAPKAPK3 and MAPKAPK5 are predicted as the major kinases in vitreous of iERM. Conclusion and clinical relevance: Our results screened candidate proteins for the treatment of fibrosis in iERM, among which tenascin-C, galectin-3-binding protein, glucose-6-phosphate isomerase, neuroserpin, collagen alpha-1(XI) chain and collagen alpha-1(II) chain are expected to become potential biomarkers or therapeutic targets for iERM. In addition, this article expand the data of vitreous phosphoproteome, and provide a reference resource and new enlightenment for studying the pathogenic mechanisms of retinal diseases.

Project description:GLP-1 induces an increase in heart rate, but its mechanism remains elusive. We used a phosphoproteomic approach to evaluate the effect of GLP-1 on the porcine sinus node. Tissue samples from the sinus node region and left ventricle were collected from eight animals following a 60-minute infusion with either GLP-1 or saline. We employed a tandem mass tag (TMT) labeling strategy to multiplex samples, enriched for phosphorylated peptides by titanium dioxide chromatography, and pre-fractionated the resulting peptide pool prior to LC-MS/MS measurements on an Orbitrap Ascend Tribrid mass spectrometer.
