Proteome analysis related to unsaturated fatty acid synthesis by interfering bovine adipocyte ACSL1 gene

ABSTRACT: Unsaturated fatty acids (UFAs) in beef are essential for human health. Long-chain fatty acyl-CoA synthase 1 (ACSL1) is a crucial gene for UFAs synthesis in bovine adipocytes. To assess the protein expression profile during UFAs synthesis, we performed a proteomic analysis of bovine adipocytes by RNA interference and non-interference with ACSL1 using label-free techniques.

Project description:Bovine preadipocytes were isolated from subcutaneous fatty tissue and induced differentiation into adipocytes and RNAs were extracted from preadipocytes and adipocytes respectively. Small RNA-seq was performed by Beijing Genomics Institute biotechnology. A total of 250 differential expression miRNAs were screened, while 131 miRNAs were highly expressed in bovine adipocytes and 119 miRNAs were highly expressed in bovine preadipocytes. KEGG pathway analysis presented that 4.76% (p-value<0.001) differently expression genes enriched in lipid metabolism. GO analysis showed the target genes were mainly associated with cell process, cell and binding. Together these results provided important insights into the research of miRNAs for bovine preadipocytes differentiation.

Project description:The potential for dietary supplementation with n-3 polyunsaturated fatty acids (n-3 PUFA) to improve reproductive efficiency in cattle has received much interest. The mechanisms by which n-3 PUFA may affect physiological and biochemical processes in key reproductive tissues are likely to be mediated by significant alterations in gene expression. We used microarrays to assess endometrial gene expression on day 17 of the estrous cycle in n-3 PUFA compared with control fed heifers. Beef heifers were supplemented with a rumen protected source of either a saturated fatty acid (CON; palmitic acid) or high n-3 PUFA (n-3 PUFA; 275 g) diet per animal per day for 45 days and global gene expression was determined in uterine endometrial tissue using an Affymetrix® oligonucleotide bovine array.

Transcriptome Analysis of a Bovine Mammary Epithelial Cell Model with Different Milk Fat Synthesis Capacity

Project description:Synthesis of milk fat is a complex biochemical process regulated by a series of molecular events. To determine gene expression changes during milk fat synthesis in bovine mammary epithelial cells, we established a cell model with a high capacity for milk fat synthesis using stimulation with acetate and β-hydroxybutyrate. RNA sequencing was used to identify differentially expressed genes (DEGs) between the high-milk-fat and control cells. A total of 625 DEGs (358 upregulated, 267 downregulated) were identified. Among the highly expressed genes, there was enrichment for terms associated with fatty acid synthesis, activation, and triacylglycerol synthesis, consistent with active milk fat synthesis. Kyoto Encyclopedia of Genes and Genomes analysis suggested that DEGs were most enriched in the “lipid metabolism” subcategory of the “metabolism” category, and in the “signal transduction” subcategory of the “environmental information processing” category. Although an in vitro cell model cannot completely simulate in vivo lactation, it eliminates interference from other cell types and from the synthesis of other milk components during transcriptome profile analysis. This work provides a profile of gene expression changes that occur during milk fat synthesis in bovine mammary epithelial cells, which furthers our understanding of the molecular regulation of lipid metabolism.

Sortilin-mediated translocation of mitochondrial ACSL1 impairs adipocyte thermogenesis and energy expenditure in male mice

Project description:Beige fat activation involves a fuel switch to fatty acid oxidation following chronic cold adaptation. Mitochondrial acyl-CoA synthetase long-chain family member 1 (ACSL1) localizes in the mitochondria and plays a key role in fatty acid oxidation; however, the regulatory mechanism of the subcellular localization remains poorly understood. Here, we identify an endosomal trafficking component sortilin (encoded by Sort1) in adipose tissues that shows dynamic expression during beige fat activation and facilitates the translocation of ACSL1 from the mitochondria to the endolysosomal pathway for degradation. Depletion of sortilin in adipocytes results in an increase of mitochondrial ACSL1 and the activation of AMPK/PGC1α signaling, thereby activating beige fat and preventing high-fat diet (HFD)-induced obesity and insulin resistance. Collectively, our findings indicate that sortilin controls adipose tissue fatty acid oxidation by substrate fuel selection during beige activation and provides a potentialnovel targeted approach for the treatment of metabolic diseases.
