Project description:Magnetic field therapy holds promise for tumor treatment, yet its underlying mechanisms remain controversial. The traditional ROS theory fails to fully explain the observed magnetic field-induced inhibition. Building on prior findings, we identified the key target of magnetic field-mediated tumor suppression and established a magnetic inhibition model. Integrative multi-omics analysis (transcriptome + proteome) of 293T and A549 cells exposed to (5 mT, 20 Hz) magnetic fields pinpointed RSRP1 as a critical molecular target. We experimentally validated RSRP1's essential role using CRISPR-Cas9-mediated gene disruption and TET-on inducible expression systems. Proteomic analysis revealed that magnetic field exposure (5 mT, 20 Hz) induced >200-fold upregulation of RSRP1 protein in both non-tumorigenic 293T and tumorigenic A549 cells. CRISPR-Cas9-mediated RSRP1 knockout completely abolished A549 cells' responsiveness to magnetic fields. Notably, restoring RSRP1 expression via a doxycycline-inducible system reinstated magnetic field-induced secretion of inhibitory factors, with conditioned media from exposed cells significantly inhibiting naive tumor cell proliferation (p<0.05). Magnetic field (5 mT, 20 Hz) exposure transcriptionally upregulated RSRP1 endogenously, as confirmed by transcriptomic and qPCR analyses, while exogenously overexpressed RSRP1 via a doxycycline-inducible system remained unresponsive, indicating that magnetic regulation requires endogenous transcriptional control. This study establishes a "magnetic field→RSRP1→secretory factor→target response" cascade, providing a transformative framework for non-invasive tumor therapy.

Project description:Using diamagnetic levitation, we have exposed A. thaliana in vitro callus cultures to five environments with different levels of effective gravity (from levitation i.e. simulated mg* to 2g*) and magnetic fields (10.1 to 16.5 Tesla) and we have compared the results with those of similar experiments done in a Random Position Machine (simulated micro g) and a Large Diameter Centrifuge (2g) free of high magnetic fields. Microarray analysis indicates that there are changes in overall gene expression of the cultured cells exposed to these unusual environments but also that gravitational and magnetic field produce synergic variations in the steady state of the transcriptional profile of A. thaliana. Significant changes in the expression of structural, abiotic stress and secondary metabolism genes were observed into the magnet field. These results confirm that the strong magnetic field, both at micro g* or 2g*, has a significant effect on the expression of these genes but subtle gravitational effects are still observable. These subtle responses to microgravity environments are opposite to the ones observed in a hypergravity one.

Project description:The biological effects of magnetic field (MF) have been studied and applied in medicine over the past four decades. However, high-intensity pulse magnetic field (HI-PMF), which may exert stronger biological effects in a shorter time, is rarely studied. Herein, this study aims to investigate the biological effects of 2.5 T HI-PMF on Escherichia coli and the consequent physiological alterations. After being treated by HI-PMF, a notable increase in intracellular NADH/NAD+ ratio was observed, accompanied by an enhanced survival rate. Transcriptome analysis revealed significant changes in genes associated with glycolysis, TCA cycle and oxidative phosphorylation, suggesting that HI-PMF induces an increase in glucose metabolism. Interestingly, the decreased intracellular glucose concentration could alleviate the inhibition of cell division, which was identified as the reason for the increased survival rate after being treated by HI-PMF. Conversely, low intracellular glucose level led to cell death under HI-PMF. Therefore, the results suggest that HI-PMF treatment promotes cellular glucose metabolism and further influences cell survival depending on intracellular glucose concentrations.
