MCF7 breast cancer cells treated with mesenchymal stem cell-derived exosomes

Project description:Human mesenchymal stem cell (MSC)-conditioned medium (CM) was previously reported to affect the biology of tumor cells; however, the precise mechanisms remain unclear. Here, we show that MSCs secreted 40- to 100-nm particles, which have the typical characteristics of exosomes, and these MSC-derived exosomes promoted migration of the breast cancer cell line MCF7. To further investigate the effect of MSC-exosomes on MCF7, we analyzed the gene expression profiles of MCF7 treated with or without MSC-exosomes for 24 h. Investigation of whole genome gene expression level changes in breast cancer cell line MCF7 which were treated with or without mesenchymal stem cell-derived exosomes. This study uses total RNA recovered from two samples. One sample is MCF7 treated with PBS for 24 hours and another one is MCF7 treated with mesenchymal stem cell-derived exosomes for 24hours. The ultimate concentration of mesenchymal stem cell-derived exosomes used in this experiment was 400ng/ul.

diaPASEF analysis of exosomes from stem cells

Project description:we used highly sensitive mass spectrometry diaPASEF (parallel accumulation serial fragmentation combined with the data-independent acquisition)27 to quantify proteins of exosomes derived from umbilical cord mesenchymal stem cells (UCMSCs) and rat bone marrow stem cells (BMSCs). 4206 human proteins and 5341 rat proteins were detected from UCMSC exosomes and BMSC exosomes, respectively. From these proteins, we identified the core proteome of exosomes which is common to UCMSCs and BMSCs. In addition, we identified two exosomal proteins which can be used as the potential marker for differentiation ability of BMSCs. In summary, the exosomal quantitative proteome presented by this study can serve as a valuable resource for further fundamental and biomarker researches of exosomes.

Single cell RNA-sequencing of induced Neural Plate Border Stem Cells

Project description:We describe a so far uncharacterized, embryonic and self-renewing Neural Plate Border Stem Cell (NBSC) population with the capacity to differentiate into central nervous and neural crest lineages. NBSCs can be obtained by neural transcription factor-mediated reprogramming (BRN2, SOX2, KLF4 and ZIC3) of human adult dermal fibroblasts and peripheral blood cells (induced Neural Plate Border Stem Cells, iNBSCs) or by directed differentiation from human induced pluripotent stem cells (NBSCs). Moreover, human (i)NBSCs share molecular and functional features with an endogenous NBSC population isolated from neural folds of E8.5 mouse embryos. Upon differentiation, iNBSCs give rise to either (1) radial glia-type stem cells, dopaminergic and serotonergic neurons, motoneurons, astrocytes, and oligodendrocytes or (2) cells from the neural crest lineage. Here we provide single cell RNA-sequencing data of six iNBSC lines (310 cells total). iNBSCs were single-cell-sorted and RNA sequencing was performed following the Smart-seq2 protocol. This dataset further supports the notion that iNBSC cultures mainly consist of stem cells with a molecular and functional neural plate border-like identity and a minor fraction of cells that show signs of some spontaneous differentiation towards sensory neurons.

Screening for the key miRNAs of exosomes that promote tumor progression and metastasis

Project description:Intercellular communication is crucial for breast cancer progression and metastasis. Yet, the role of cancer-derived exosomes and their specific miRNAs cargo in mediating intercellular communications needed to be further elucidated. MiRNAs control gene expression post-transcriptionally and are selectively packaged into cancer-derived exosomes. Therefore, we hypothesized that miRNAs in cancer-derived exosomes were most likely the essential components of intercellular communication. To identify the key miRNAs, we chose metastatic MDA-MB-231 and non-metastatic MCF-7 for comparison. The miRNAs profiling of exosomes derived from these two kinds of cells as well as the endogenous miRNAs profiling of the cells were conducted by microarrays.
