Proteome of extracellular vesicles derived from breast cancer cells

ABSTRACT: MDA-MB-231 cells were irradiated with 10 Gy X-rays and the medium was harvest for extracellular vesicles isolation 24 hours post-irradiation. Proteins carrying by EVs of irradiated and control cells were then analysed by LC-MS.

Radiotherapy reduces the risk of cancer recurrence and death, but the fact that it's accompanied by multiple side effects including muscle fibrosis and weakness, seriously affects the life quality of patients. RNA-seq of gastrocnemius (GA) muscle tissue from mice treated with extracellular vesicles (EV) from MCF-10A or MDA-MB-231 cells

Project description:To compare gene expression changes in skeletal muscle caused by EVs from normal (MCF-10A) and cancer (MDA-MB-231) cells, we analyzed RNA isolated from the GA muscle of EV-treated female NOD scid gamma (NSG) mice. Mice had received tail-vein injections of EVs twice a week for 5 weeks (~10 ug EV per injection). Gene expression in muscle from mice treated with MDA-MB-231-derived EVs was compared to mice treated with MCF-10A-derived EVs.

Characterization of tumor extracellular vesicle RNA cargo

Project description:Comparative RNA profiling between tumor cells and their secreted extracellular vesicles. Results revealed enrichment in genes involved in cellular migration and metastasis in extracellular vesicles, in agreement with their role as mediators of tumor progression. Mice were orthotoplically transplanted with MDA-MB-231 Breast Adenocarcinoma cells. Cells and extracellular vesicles (EVs) from the resulting tumors were isolated. EVs were characterized by electron microscopy and Nanoparticle Tracking Analysis before total RNA isolation for comparative analysis with cellular RNA. Three biological replicates were analyzed in (technical) duplicate.

Rapid isolation of extracellular vesicles from cell culture and biological fluids using a synthetic peptide with specific affinity for heat shock proteins

Project description:Background: Exosomes and extracellular vesicles (EVs) are increasingly recognized as important sources of biomarkers for disease study and diagnosis. Results: A synthetic peptide, Vn96, allows for capture of EVs from biological fluids using basic laboratory equipment. Conclusion: The Vn96-captured EVs are qualitatively equivalent or superior to exosomes isolated by ultracentrifugation. Significance: The Vn96 peptide provides an effective affinity-capture method for the isolation of EVs from biological fluids. In order to compare different methods of exosome purification, we compared RNA content of exosomes purified with each method. We used two different breast cancer cell lines MCF7 and MDA-MB-231. Extracellular vesicles induce aggressive phenotype of luminal breast cancer cells by PKM2 phosphorylation

Project description:We report that EVs originated from cancer cells can modulate glucose metabolism in the recipient cancer cells and induce cell proliferation and aggressive phenotype. Two breast cancer cell lines with different levels of glycolytic activity, MDA-MB-231 of a claudin low-type breast cancer cell and MCF7 of luminal type breast cancer cell, were selected and co-cultured, as the originating and recipient cells using indirect co-culture system such as transwell system or microfluidic system. Proteomic profiling of the co-cultured MCF7 cells revealed proliferation and dedifferentiation of the MCF7 cells following co-culture with the MDA-MB-231 cells. Transcriptomic analysis demonstrated that glycolysis increased in the co-cultured MCF7 cells, and the component analysis of glycolysis-related genes revealed that the second-most component after cytoplasm was extracellular exosomes. In addition, 36 significant KEGG pathways were identified in a total of 856 proteins identified by proteomin analysis of MDA-MB-231-induced EVs. Among these pathways were the main pathways (glycolysis/gluconeogenesis, pyruvate metabolism, and PI3K-Akt signaling pathways) that could explain the metabolic modulation of MCF7 cells. In our work, we indirectly show that it is MDA-MB-231-derived EVs that play an important role in this phenomenon. Our study highlights the potential effects of aggressive cancer cells on other surrounding cancer cells through EVs.

Project description:We report the gene expression levels in MDA-MB-231 cells on treatment with (5-oxo-2-dibenzothienylmethyl)triphenylphosphonium (1-SO) and (2-dibenylthienylmethyl)triphenylphosphonium (1-S) irradiated with UV-A light. These gene expression levels were then compared to control conditions where the cells were treated with the compounds but were not irradiated with UV-A light. The samples were additionally compared to the cells treated with 1% DMSO in DMEM media (with and without UV-A irradiation). Results: From the first set of experiments, 30 minutes of UV-A irradiation upregulated 3 and downregulated 5 genes compared to cells with no UV-exposure. The experiments also indicated that 1-SO in UV-A irradiation significantly affected gene expression whereas 1-SO without UV-A did not have a notable effect on expression levels. However, 1-S affected gene expression levels significantly with or without UV-A irradiation.
