Project description:We designed this experiment to investigate the transcriptional changes in gonads as a result of sex transformation. Here we performed transcriptional profiling of the ovary transformed into testis from the tra loss of function (XX_tra_lof), testis transformed into ovary from the tra gain of function (XY_tra_gof) and ovary transformed into testis in dsxM gain of function (XX_DsxM_gof/lof) Drosophila melanogaster third instar larvae in biological quadruplicates. In addition, as controls we sequenced ovaries and testes from the female and male wildtype larvae respectively. We constructed polyA+ libraries of the gonads, cleaned off the fatbody and performed 50 bp, stranded single-end RNA-Seq.

Project description:Geoduck clams (Panopea generosa) were collected from Puget Sound, WA in November, 2014. Male and female geoduck gonads were sampled at three reproductive stages over the course of three months: early, middle, and late. Early stage indicates that the gonad cells are just beginning to differentiate and late stage indicates that the geoduck are ready to spawn. The goal of this study was to identify biomarkers of the geoduck reproductive cycle. Due to the restriction of our current data model we can not add multiple names as Lab Head. However, this Project has been equally led by Steven B. Roberts and Brook L. Nunn.

Project description:The main goal of our study is to identify the molecular events that determine the gonadal identity in mammals. Although testis and ovary arise from a common embryonic primordium, they represent outcomes of opposing fate determination. This decision to differentiate into a testis or an ovary hinges upon the balance between two antagonizing factors, pro-testis SOX9 and pro-ovary β-catenin. This microarray analysis led to the identification of the genes involved in the fate of XX and XY gonads in absence of SOX9 and beta-catenin We developed mouse genetic models that lack either Sox9, β-catenin, or both specifically in the somatic cells. All embryos used in this study resulted from the crossing between Ctnnb1f/f; Sox9f/f females with Sf1-cre+/Tg ; Ctnnb1+/f; Sox9+/f males. XX and XY fetal gonads were collected at embryonic day E14.5

Single nucleotide insertion within a remote enhancer causes XX mice to be born as males

Project description:In this study, we show that mice carrying a 3 bp deletion or 1 bp insertion within the SOX9 BS of Enh13 are born as XX males. At adult stages, mice appear externally and internally as males but their testes are small and devoid of sperm due to the lack of the Y chromosomes. At embryonic stages, XX homozygous gonads appear as ovotestis at various severities with the 1 bp insertion appearing more as a testis. Transcriptomic analysis of E12.5 gonads shows that indeed the XX homozygous gonads are positioned in between the testis and ovary while XY homozygous gonads are indistinguishable of wild type (WT) XY gonads. These findings implicate Enh13 as a dual regulatory element essential for both testicular and ovarian development, placing it at the critical intersection of sexual differentiation pathways.

The embryonic ontogeny of the gonadal somatic cells in mice and monkeys [sc3seq]

Project description:In the early fetal stage, the gonads are bipotent and only later become the ovary or testis, depending on the genetic sex. Despite many studies examining how sex determination occurs from biopotential gonads, the spatial and temporal organization of bipotential gonads and their progenitors is poorly understood. Here, using lineage tracing in mice, we find that the gonads originate from a T+ primitive streak through WT1+ posterior intermediate mesoderm and appear to share origins anteriorly with the adrenal glands and posteriorly with the metanephric mesenchyme. Comparative single cell transcriptomic analyses in mouse and cynomolgus monkey embryos reveals the convergence of the lineage trajectory and genetic programs accompanying the specification of biopotential gonadal progenitor cells. This process involves sustained expression of epithelial genes and upregulation of mesenchymal genes, thereby conferring a unique epithelial/mesenchymal hybrid state. Our study provides key resources for understanding early gonadogenesis in mice and primates.
