Project description:Using high-throughput RNA sequencing, we developed a spatiotemporal transcriptome atlas for seed development of eight maize inbred lines based on 144 samples from the middle to late stages of grain development. A total of 26,747 genes with FPKM value more than 1 at least one sample were found to be involved in programming grain development. Global comparisons of genes expression highlighted the fundamental transcriptomic reprogramming and the phases of development. Coexpression analysis provided further insight into the dynamic reprogramming of the transcriptome by revealing functional transitions during maturation. Combined with grain moisture content and grain dehydration rate of different developmental time points of eight maize inbred lines, we captured a large number of genes related to grain moisture content and grain dehydration rate, which should help elucidate key mechanisms and regulatory networks that underlie grain dehydration during maize grain development. These results provide a comprehensive understanding of which biological processes are involved in the regulation of moisture variety of maize grain, the general principles of which provide a new perspective on improving maize grain dehydration characteristics. Meanwhile, this study provides a valuable resource for understanding the genetic regulation of maize grain development.

Project description:Grain filling in maize (Zea mays L.) is intricately linked to cell development, involving the regulation of genes responsible for the biosynthesis of storage reserves (starch, proteins, and lipids) and phytohormones. However, the regulatory network coordinating these biological functions remains unclear. In this study, we identified 1,744 high-confidence target genes co-regulated by the transcription factors (TFs) ZmNAC128 and ZmNAC130 (ZmNAC128/130) through chromatin immunoprecipitation sequencing coupled with RNA-seq analysis in the zmnac128/130 loss-of-function mutants. We further constructed a hierarchical regulatory network using DNA affinity purification sequencing analysis of downstream TFs regulated by ZmNAC128/130. In addition to target genes involved in the biosynthesis of starch and zeins, we discovered novel target genes of ZmNAC128/130 involved in the biosynthesis of lipids and indole-3-acetic acid (IAA). Consistently, the number of oil bodies, as well as the contents of triacylglycerol and IAA were significantly reduced in zmnac128/130. The hierarchical regulatory network centered by ZmNAC128/130 revealed a significant overlap between the direct target genes of ZmNAC128/130 and their downstream TFs, particularly in regulating the biosynthesis of storage reserves and IAA. Our results indicated that the biosynthesis of storage reserves and IAA is coordinated by a multi-TFs hierarchical regulatory network in maize endosperm.

Project description:The orchestration of source, flow, and sink is fundamental to efficient grain filling in maize, yet the regulatory network synchronizing these processes remains a major enigma. To identify key regulators, we used the sucrose transporter ZmSUT1, a critical component of the ‘flow’ pathway, as a molecular target. This approach led to the discovery of ZmbZIP39 that acts as a central coordinator of grain filling. We demonstrate that ZmbZIP39 governs the entire process by simultaneously regulating photosynthesis (source), sucrose transport (flow), and sucrose metabolism and nutrient storage (sink). Mechanistically, ZmbZIP39 directly activates the expression of ZmSUT1 and ZmSUT7, facilitating sucrose loading and long-distance transport from leaves to developing kernels. It also promotes sucrose cleavage by directly upregulating three sucrose synthase-encoding genes. Furthermore, ZmbZIP39 directly controls the expression of the zein-encoding genes and endosperm-filling hub Opaque2, thereby driving nutrient storage and endosperm development. ZmbZIP39 also functions within a higher-order regulatory complex, synergizing with key factors like ZmABI19, ZmbZIP29, OHP1, OHP2, and ZmNAC130, to amplify their collective impact. ZmbZIP39-mediated regulatory capacity is further enhanced by its autoactivation activity, creating a positive feedback loop that robustly strengthens its downstream network. Our findings establish ZmbZIP39 as a master regulator that optimizes source-flow-sink relations.¬
