Investigation of spontaneous pelvic organ prolapse in the rhesus macaque model

Project description:An appropriate model is urgently required for pathological mechanism investigation of pelvic organ prolapse and the development of new therapeutic strategies. It is generally thought that damage to the vagina and its supportive tissues due to vaginal birth, menopause, and so on can result in loss of vaginal support and prolapse of the pelvic organs. Currently, ovariectomy and simulated vaginal delivery in animal models are applied to mimic the status of menopause and induce vaginal injury, however, the complex biochemical and biomechanical changes induced by vaginal delivery and prolapse is difficultly achieved by these methods. The rhesus macaque shares many features similarly to women, such as pelvic floor anatomy and parturition, and makes an excellent experimental model for the study of prolapse. Besides, the rhesus macaque develops prolapse spontaneously. Thus, there has great value to investigate the changes of spontaneous prolapse and make them animal models for the study of prolapse. In this study, we will assess the histomorphology, biochemical property and biomechanical property of prolapsed vagina. And then, cell atlas of normal and prolapsed vagina will be constructed by single-cell transcriptome sequencing. We will reveal the cell type specific molecular mechanisms. This study will improve the understanding of spontaneous prolapse in the rhesus macaque and provide fundamental data for the application of spontaneous prolapse as animal models to study POP.

Patient-derived organoids of pancreatic ductal adenocarcinoma for subtype determination and clinical outcome prediction

Project description:Two molecular subtypes of pancreatic ductal adenocarcinoma (PDAC) have been proposed: the “Classical” and “Basal-like” subtypes. However, the “molecular” classification has not been applied in real-world clinical practice. This study aimed to establish patient-derived organoids (PDOs) for PDAC and evaluate their application in subtype classification and clinical outcome prediction. We constructed a PDO library for morphologic and RNA-seq analyses and drug response assays in vitro. PDOs of PDAC were established at a high efficiency (> 70%) with at least 100,000 live cells. Morphologically, PDOs were classified as gland-like structures (GL type) or densely proliferating inside (DP type). RNA-seq analysis revealed that the “morphological” subtype (GL vs. DP) roughly corresponded to the “molecular” subtype (“Classical” vs. “Basal-like”). The proposed “morphological” classification predicted clinical treatment response and prognosis; the median overall survival of patients with GL type were significantly longer than that of those with DP type (P < 0.005). The GL type showed a better response to gemcitabine than the DP type in vitro, whereas the drug response of the DP type was improved by the combination of ERK and autophagy inhibition. PDAC PDOs facilitated subtype determination and clinical outcome prediction, thereby advancing precision medicine for PDAC.

Functionally relevant prediction model for colorectal cancer

Project description:Filtered selection coupled with support vector machines generate functionally relevant prediction model for colorectal cancer. In this study, we built a model that uses Support Vector Machine (SVM) to classify cancer and normal samples using Affymetrix exon microarray data obtained from 90 samples of 48 patients diagnosed with CRC. From the 22,011 genes, we selected the 20, 30, 50, 100, 200, 300 and 500 genes most relevant to CRC using the Minimum-Redundancy–Maximum-Relevance (mRMR) technique. With these gene sets, an SVM model was designed using four different kernel types (linear, polynomial, radial basis function and sigmoid).

Project description:The nucleotide reverse transcriptase inhibitor (NRTI) tenofovir has been extensively tested as a topical preventative against vaginal and rectal HIV transmission. We sought to determine how use of this product affects gene expression. We used samples from MTN-014, a study of vaginal and rectal gel application. We obtained vaginal biopsies before treatment initiation and after two weeks of daily use of tenofovir 1% gel applied rectally or vaginally. We isolated RNA from vaginal biopsies preserved with RNAlater. Gene expression was measured from all three samples (three time points) from each participant.
