Project description:Col-0 transgenic plants expressing MSH1P::FLAG::RPL18 were generated in both msh1 homozygous mutant (SAIL-877-F01) and wild type backgrounds with cloned MSH1 sequence described in Xu et al. (2011) and with transformation by the floral dip method (Clough and Bent, 1998). Positive transgenic lines were screened for FLAG tag by immunoblotting methods. Polysomal mRNA from MSH1-specific cells was obtained by ribosome purification using the TRAP method essentially as reported by (Reynoso et al., 2015) from floral stem tissues.

Project description:Anti-FLAG-affinity purification of Umbrea associated factors from nuclear extracts of Drosophila Schneider cells expressing FLAG-HA-Umbrea. Anti-FLAG -affinity purifications from Schneider cell nuclear extracts not expressing any FLAG-tagged protein served as a control SDS-PAGE separting both samples, 9 slices per sample, reduction and alkylation, digestion with trypsin. LC-MS on a Ultimate3000-LTQ Orbitrap XL Raw data were analysed using the MaxQuant 1.2.2.5 software package. Identified proteins were considered as interaction partners if their MaxQuant intensities displayed a greater than 10 fold enrichment compared to control anti-FLAG purifications from L2-4 nuclear extracts not expressing any FLAG-tagged protein

Only the GFP-positive cells were used for study and expanded in DMEM medium.

Project description:In order to assess Tet1 binding, we first generated a Flag tagged Tet1 ES cells and then knocked out Dnmt3a in the [WT, Tet1-Flag] cells. By Tet1 ChIP and Flag ChIP, we showed that Tet1 binding was complementary to Dnmt3a. And Tet1 binding was not affected or slightly increased at majority of its targets.
