Prostate cancer (PCa) is the most common malignant tumor in men. Currently, there are few prognosis indicators for predicting PCa outcomes and guiding treatments. Here, we perform comprehensive proteomic profiling of 918 tissue specimens from 306 Chinese patients with PCa using data-independent acquisition mass spectrometry (DIA-MS). Project description:Prostate cancer (PCa) is a non-cutaneous malignancy that is the most frequent cancer found in males worldwide, and its mortality rate is increasing every year. Although, PCa grows slowly and is not a threat to survival, advanced PCa, metastatic PCa, is aggressive form of PCa and can spread to other organs, resulting in the 5-year survival rate dropping to 30%. However, there are no molecular markers for advanced and/or aggressive PCa, and it is an urgent clinically need to find biomarkers for prognosis and prediction of advanced PCa. Here, we used mass spectrometry-based proteomics to discover new biomarkers for prognosis and prediction of advanced PCa in tissue obtained from PCa patients who were diagnosed with T2, T3 and metastasis in regional lymph nodes. We identified 1,904 proteins and 1,673 proteins were quantified from PCa tissues, respectively. In particular, 344 differentially expressed proteins (DEP) of which 124 were up-regulated and 216 were down-regulated were listed. From PLS-DA score plotting using SIMCA P+ software and GO and KEGG enrichment analysis by DAVID, we also propose SRM, NOLC1, and PTGIS, which we found through this proteomics analysis, as representing new protein biomarkers for diagnosis of advanced PCa. The proteomics results were verified by immunoblotting assay in metastatic PCa cell-lines and indirect ELISA in prostate specimens. In particular, SRM is significantly increased depending on cancer progress stages, so the possibility of biomarker for prognosis and prediction of advanced PCa is confirmed.

Project description:The purpose of this study are: - To identify new biomarkers specific for prostate cancer (PCa) that can be used as diagnostic markers in the urine of individuals with high probability of PCa (abnormal PSA and/or digital rectal examination). - To validate the utility of these new biomarkers, as well as others already known such as PCA3, fusion gene TMPRSS2-ERG, GOLPH2 and SPINK1. - To establish a prediction model for the diagnosis of PCa based on the expression of these biomarkers. - To validate this model within the framework of an opportunist programme of early diagnosis. - Within the framework of this programme, to associate a series of social-demographic, antropometric, life-style and occupational variables for establishing a risk model predictor that could be associated with the model based on the biomarkers. Methodology: The study is divided in three phases based on three different cohorts of patients: -Phase 1. Identification of biomarkers. This phase includes 60 patients (10 normal prostates and 50 PCa). Form these tumors a screening of miRNAs will be performed. -Phase 2. Validation of the new biomarkers and others already known by means qRT-PCR on urine samples from 300 patients (200 with histological diagnosis of PCa and 100 with an histological negative result). -Phase 3. Prospective validation in a prospective cohort iof 1065 patients included in an opportunist programme of early detection of PCa.

Project description:: Prostate cancer (PCa) is the most commonly diagnosed genital cancer in men worldwide. Among patients who developed advanced PCa, 80% suffered from bone metastasis, with a sharp drop in the survival rate. Using quantitative global succinylome analysis, we characterized a significant increase of lysine 118 succinylation (K118su) of lactate dehydrogenase A (LDHA), which plays a role in increasing LDHA activity. As a substrate of SIRT5, LDHA-K118su significantly increased the migration and invasion of PCa cells and LDHA activity in PCa patients.

Project description:Purpose: Prostate-specific membrane antigen-targeted radioligand therapy (PSMA-RLT) is effective against prostate cancer (PCa), but all patients relapse eventually. Poor understanding of the underlying resistance mechanisms represents a key barrier to development of more effective RLT. We investigate the proteome and phosphoproteome in a mouse model of PCa to identify signaling adaptations triggered by PSMA-RLT.
Experimental Design: Therapeutic efficacy of PSMA-RLT was assessed by tumor volume measurements, time to progression, and survival in C4-2 or C4-2 TP53-knockout tumor-bearing Nod scid gamma mice. Two days post-RLT, the (phospho)proteome was analyzed by mass spectrometry.
Results: PSMA-RLT significantly improved disease control in a dose-dependent manner. (Phospho)proteomic datasets revealed activation of genotoxic stress response pathways, including deregulation of DNA damage/replication stress response, TP53, androgen receptor, PI3K/AKT, and MYC signaling. C4-2 TP53-knockout tumors were less sensitive to PSMA-RLT than parental counterparts, supporting a role for TP53 in mediating RLT responsiveness.
Conclusions: We identified signaling alterations that may mediate resistance to PSMA-RLT in a PCa mouse model. Our data enable the development of rational synergistic RLT-combination therapies to improve outcomes for PCa patients.

Project description:Patients with oral preneoplastic lesion (OPL) have high risk of developing oral cancer. Although certain risk factors such as smoking status and histology are known, our ability to predict oral cancer risk remains poor. The study objective was to determine the value of gene expression profiling in predicting oral cancer development. Gene expression profile was measured in 86 of 162 OPL patients who were enrolled in a clinical chemoprevention trial that used the incidence of oral cancer development as a prespecified endpoint. The median follow-up time was 6.08 years and 35 of the 86 patients developed oral cancer over the course. Gene expression profiles were associated with oral cancer-free survival and used to develope multivariate predictive models for oral cancer prediction. We developed a 29-transcript predictive model which showed marked improvement in terms of prediction accuracy (with 8% predicting error rate) over the models using previously known clinico-pathological risk factors. Based on the gene expression profile data, we also identified 2182 transcripts significantly associated with oral cancer risk associated genes (P-value<0.01, single variate Cox proportional hazards model). Functional pathway analysis revealed proteasome machinery, MYC, and ribosomes components as the top gene sets associated with oral cancer risk. In multiple independent datasets, the expression profiles of the genes can differentiate head and neck cancer from normal mucosa. Our results show that gene expression profiles may improve the prediction of oral cancer risk in OPL patients and the significant genes identified may serve as potential targets for oral cancer chemoprevention. Project description:Prostate cancer (PCa) is one of the most prevalent cancer types in men worldwide. However, the main diagnostic tests available for PCa have limitations and need biopsy for histopathological confirmation of the disease. The prostate-specific antigen (PSA) is the main biomarker used for PCa early detection, but an elevated serum concentration is not cancer-specific. Therefore, there is a need for discovery of new non-invasive biomarkers that can accurately diagnose PCa. Here, we used trichloroacetic acid-induced protein precipitation and liquid chromatography-mass spectrometry to profile endogenous peptides in urine samples from patients with PCa (n = 33), benign prostatic hyperplasia (n = 25), and healthy individuals (n = 28). Receiver operating characteristic (ROC) curves were performed to evaluate the diagnostic performance of urinary peptides. In addition, proteasix tool was used for in silico prediction of protease cleavage sites. We found five urinary peptides derived from uromodulin significantly altered between the study groups, all of which were less abundant in the PCa group. In addition, urinary peptides outperformed PSA in discriminating between malignant and benign prostate conditions (AUC = 0.847), showing high sensitivity (81.82%) and specificity (88%). Overall, our study allowed the identification of urinary peptides with potential for use as non-invasive biomarkers in PCa diagnosis.

Project description:Prostate cancer (PCa) is a major public health issue in industrialized countries, mainly because of patients’ relapse by castration-refractory disease after androgen ablation. PCa progresses through a series of clinical states characterized by the extent of the disease, the hormonal status (castration-sensitive or castration-resistant) and the presence or absence of metastases. Progression to castration-resistant prostate cancer (CRPCa) involves several mechanisms such as ligand-independent androgen receptor activation and adaptive up-regulation of anti-apoptotic genes. CRPCa is highly aggressive and incurable, jeopardizing the patient’s quality of life and lifespan. Identifying the molecular events responsible for the progression to CRPCa is essential to avoid its development and design specific therapies. Despite the existing treatment guidelines for PCa and novel clinical trials for CRPCa, major challenges still remain for identifying specific CRPCa biomarkers and therapeutic targets for effective tailored therapy design for these patients. In this context, molecular profiling of human PCa may importantly contribute to the identification of new disease-specific biomarkers in order to improve PCa early diagnosis, prognosis and disease course and to predict strategies that help develop novel therapies. System-wide approaches such as transcriptomics and proteomics are nowadays applied to monitor molecular variations at the cellular level. Proteomics approaches offer a great potential for the discovery of novel biomarkers and the identification of new therapeutic agents by accurate quantification of proteins. Several studies have been published in the past where different proteomics approaches were used to identify PCa proteome. In the present study we analyzed the PCa proteome from several PCa cell lines representing different hormonal status of the disease to identify potential biomarkers differentially expressed in CRPCa.

Project description:Prostate cancer (PCa) is one of the most prevalent cancers among men and the fifth leading cause of cancer-related death in men. Currently, PCa suspicion is based on abnormal digital rectal examination and/or raised PSA serum levels, with prostate needle biopsy being required for a definitive diagnosis. Histopathological classification of tumours based on GS grading, cancer staging and PSA levels are used to predict the indolent or aggressive progression of the tumour as well as the likelihood of disease recurrence. These diagnosis and prognosis tools for PCa have revealed limited usefulness, especially PSA testing, which despite organ-specificity is not cancer-specific, being associated with low specificity. In this vein, new markers have been proposed in order to increase the accuracy of PCa detection, most of them proteins. Despite the significant efforts that have been undertaken for discovering other biomarkers for PCa management, few were translated into clinical practice. The simple and non-invasive nature of urine collection along with its proteome stability and storing many secreted proteins of prostate origin, makes the identification of PCa urinary biomarkers an attractive approach. With this in mind, we aimed to compare the urinary proteome profile of PCa patients with non-cancer patients in order to identify non-invasive candidate biomarkers for PCa prediction. To fulfil this task, we followed a shotgun LC-MS/MS approach using an Orbitrap instrument. A combination of two different software packages, MaxQuant and Proteome Discover was used to increase the robustness of analysis and enhance the search for new biomarkers. Proteins quantification was based on a label-free quantification approach (false discovery rate (FDR) 1%). The present dataset was used to disclosure potential markers in PCa management.

Project description:Background: Prostate cancer (PCa) and Type 2 diabetes (T2D) are two major health risks that often occur concurrently in men. Studies predict that diabetic PCa patients have a reduced risk of PCa progression, while other studies show contradicting data. This impedes unraveling the connection between PCa with T2D. Besides other drugs, Peroxisome Proliferator-Activated Receptor (PPAR) agonists have been applied as T2D medication in patients. In publicly available patient datasets high PPARγ expression correlated with advanced PCa and worse survival outcomes. Aim: We investigated the effect of PPAR agonists Bezafibrate (PPARα), Tesaglitazar (PPARα/γ), and Pioglitazone (PPARγ) on PCa tumorigenesis using primary PCa 22RV1 and metastatic PC3 cells. To get an unbiased view on the proteome repertoire of primary and metastatic PCa cells, we analysed their proteome at basal culture conditions and following treatment with PPAR agonists Bezafibrate, Tesaglitazar, and Pioglitazone. As compared to vitality assays, we chose a relatively short (24h) treatment period, assuming that early proteome alterations might provide mechanistic insight to explain the effect of PPAR agonists on vitality and proliferation.
