Multi-omics approach identify key proteins and regulatory pathways in colorectal cancer

ABSTRACT: In order to screen out robust biomarker(s) in relation to CRC development, we here took advantage of the nontargeted proteomic technologies to comprehensively profile the characterizations of multiply types of biological specimens (including tissues and blood) from patients, and tissues from CRC mice as well. The present study aimed to identify the key regulatory pathways and proteins associated with the molecular pathogenesis of CRC.

Identify the dysregulated miRNA in human colorectal cancer tissues

Project description:Dysregulated miRNA in human colorectal cancer (CRC) were identified through comparison between 4 CRC tumors and their adjacent normal tissues by miRNA array. Histologically-confirmed CRC were included in this study. CRC tissues and paired adjacent normal tissues were obtained from the resected surgical specimens. The adjacent normal tissue is composed of normal colonic mucosa located at approximately 10 cm away from the cancer tissue. miRNA profiling of 754 human miRNAs was performed using TaqMan Human MiRNA Array Set v3.0. Quantitative real-time polymerase chain reaction (Q-PCR) was performed using Applied Biosystems 7900HT Real-Time PCR System (Applied Biosystems). Results were analyzed by the SDS RQ Manager 1.2 software (Applied Biosystems).

Transcription profling of colorectal carcinoma in order to identify potential key gene expression

Project description:In order to identify potential genes that may play an important role in progression of colorectal carcinoma, we screened and validated the global gene expression using cDNA expression array on 36 CRC tissues and compared with 24 non-cancerous colorectal tissue. We investigated the differential expression levels using cDNA microarray technique in a series of 35 CRC and 24 normal samples. FoxM1 was identified as one of the dysregulated genes and was significantly overexpressed in the tumor samples in comparison to the normal samples. 35 colorectal cancer samples versus 24 normal samples.

Gene expression profiling study to identify key markers associated with disease recurrence in colorectal cancers.

Project description:The objective of this study is to identify a gene set to predict recurrence of colorectal cancer (CRC) patients. We generated RNA-seq data of 110 primary CRC samples and identified significant genes associated with recurrence of CRC. Through diverse statistical methods including generalized linear model likelihood ratio test, significant 10 genes were identified. In the validation cohorts, a risk classifier consisting of the 10 genes was an independent risk factor in colorectal cancers.
