Sequential progenitor states mark the generation of pancreatic endocrine lineages in mice and humans

Project description:The pancreatic islet contains multiple hormone+ endocrine lineages (alpha, beta, delta, PP and epsilon cells), but the developmental processes that underlie endocrinogenesis are poorly understood. Here, we generated novel mouse lines and combined them with various genetic tools to enrich all types of hormone+ cells for well-based deep single-cell RNA sequencing (scRNA-seq), and gene coexpression networks were extracted from the generated data for the optimization of high-throughput droplet-based scRNA-seq analyses. These analyses defined an entire endocrinogenesis pathway in which different states of endocrine progenitor (EP) cells sequentially differentiate into specific endocrine lineages in mice. Subpopulations of the EP cells at the final stage (EP4-early and EP4-late) show different potentials for distinct endocrine lineages. epsilon cells and an intermediate cell population were identified as distinct progenitors that independently generate both alpha and PP cells. Single-cell analyses were also performed to delineate the human pancreatic endocrinogenesis process. Although the developmental trajectory of pancreatic lineages is generally conserved between humans and mice, clear interspecies differences, including differences in the proportions of cell types and the regulatory networks associated with the differentiation of specific lineages, have been detected. Our findings support a model in which sequential transient progenitor cell states determine the differentiation of multiple cell lineages and provide a blueprint for directing the generation of pancreatic islets in vitro.

Liver high-throughput sequencing of high-fat C57BL/6J wild-type mice

Project description:Eight weeks after C57BL / 6J mice fed a high-fat diet (HFD), the degree of insulin resistance was assessed by intraperitoneal glucose tolerance test (IPGTT) and the insulin resistance model was successfully established. Mice were then treated with resveratrol and metformin for 6 weeks and blood and liver tissue samples were collected. Blood biochemical parameters were determined by kit, protein expression of hepatic insulin signaling pathway mRNA and mRNA was determined by Western blot and real-time polymerase chain reaction, and histomorphological changes were observed by histological staining. Mouse liver tissue samples were detected by high-throughput sequencing technology to obtain differential lncRNA expression profiles. Further differentially expressed genes were analyzed using bioinformatics techniques.

Ly6C monocyte subsets transcriptome analysis from control and ApoE-/- mice

Project description:Inflammatory monocyte (MC) subset differentiation is a major feature in tissue inflammatory and atherosclerosis. The underlying molecular mechanism remains unclear. This study aims to explore molecule targets and signaling which determinate immunological features in MC subsets. ApoE-/- mice were fed a normal chow diet and switched to a high-fat (HF) diet at age 8 weeks and maintained on a HF diet for 12-14 weeks. The control C57BL/6 mice were fed a normal chow diet throughout. Blood Ly6Chigh and Ly6Clow MC subsets from control and ApoE-/- mice were isolated by flow cytometry sorting and subjected for bulk high-throughput RNA-sequencing.
