Project description:Liver tissues of Guangxi Ma chicken from 32-week old with m performance, 50-week old with high and low laying performance, and 72-week old with high and low laying performance were collected and sequenced in quadruplicate using RNA-seq. The sequences were double-ended sequenced on the DNBSEQ sequencing platform. The sequence reads were quality controlled and then aligned with genomic sequences using HISAT2 program, quantified by featureCounts program, and gene expression levels were verified by qRT-PCR with SYBR Green detection. The results will be helpful to explore the factors that affecting laying performance from the perspective of yolk synthesis and provide a theoretical basis for improving the egg production of Guangxi Ma chicken.

Project description:Laying performance is a key factor affecting production efficiency in poultry farming, but its molecular mechanism is still unclear. In this study, Yaoshan chickens, a local breed in Guizhou, China, and commercial chickens (GYR) with higher egg yield after the three-line cross improvement hybridization of Yaoshan chickens were used as animal samples. To explore the regulatory mechanism of the differences in laying performance, RNA-seq and ultra-performance liquid chromatography-tandem mass spectrometry (UPLC‒MS/MS) were used to portray the transcriptional and metabolic profiles of the ovaries of Yaoshan and GYR chickens. At the transcriptional level, 288 differentially expressed genes were upregulated in Yaoshan chickens and 353 differentially expressed genes were upregulated in GYR chickens. In addition, GSEA revealed that ECM-receptor interactions and the TGF-β signalling pathway were inhibited, resulting in increased egg production in GYR chickens. Furthermore, the upregulation of thiamine and carnitine was identified by metabolomic analysis to promote the laying performance of chickens. Finally, comprehensive analyses of the transcriptome and metabolome found that thiamine and carnitine were negatively correlated with ECM-receptor interactions and the TGF-β signalling pathway, which jointly regulate the laying performance of Yaoshan chickens and GYR chickens. In conclusion, our study delineates differences in the transcriptional and metabolic profiles of the ovaries of Yaoshan and GYR chickens during the peak egg production period and provides new hypotheses and clues for further research on poultry egg production performance and the improvement of economic benefits.

Project description:The freshwater zooplankton Daphnia magna has been used for the validation of chemical toxicity tests such as OECD Test Guideline 202 and 211. Previously, it has been demonstrated that treatment of juvenile hormones (JHs) or their analogs to mother daphnids could induce male offspring production. Based on this funding, a rapid screening method for detection of chemical with JH-activity was recently developed by using adult D. magna. This screening system can validate whether arbitrary chemical has JH-activity by investigating the male offspring inducibility. Although this short-term screening system will be possible to enhance a throughput of detection efficiency, JH-responsive genes in the ovary including developing oocytes at JH-sensitive period for male sex determination are still largely undescribed. Here, we conducted comparative microarray analyses using ovary treated by fenoxycarb (Fx; artificial JH agonist) and methyl farnesoate (MF; a putative innate JH in daphnids) to make the comprehensive gene catalogs in response to JH molecules in ovary including developing oocytes at JH-sensitive period for male sex determination. We successfully created the comprehensive ovarian gene catalogs in response to Fx and MF. Furthermore, we demonstrated that the hemoglobin genes are well-conserved JH-responsive element even in the ovary, and a potential toxic effect of JH agonist is suppression of vitellogenin gene expression that might cause reduction of offspring number. This is first report demonstrating the differential transcriptomics profiles between MF and artificial JH agonist in D. magna ovary, making it possible for understanding the tissue-specific mode-of-action of JH molecule.

Project description:Our two main aims were 1) to isolate age-related changes in gene expression in queens of the eusocial bumble bee, Bombus terrestris; and 2) to determine whether experimentally increasing the costs of reproduction (by removing eggs) caused changes in age-related gene expression in these queens. To address these aims we extracted RNA from three key tissues (brain, fat body and ovary) from queens at two time points (10% and 60% mortality phases). Each of these queens had experienced one of two treatments: an egg removal (R) treatment (to increase the costs of reproduction) and an egg removal and replacement (C) treatment (to control for the effects of disturbance caused by egg removal).

Project description:Purpose: To identify the key regulatory genes and pathways involved in chicken high egg productivon in HPG axis. Methods: A total of 856 Chinese Luhua chicken was raised in poultry breeding farm of Sichuan Agricultural University, the highest two hundred and the lowest two hundred chicken egg production were considered as high egg production (HEP) and low egg production (LEP) according to the total egg number at 300 days of age, respectively, integrated with RNA-seq sequencing of samples of HPG axis (hypothalamus, pituitary gland and ovary) from three HEP and three LEP chickens at 300 days of age. Results: A total 86.7 Gb RNA-seq sequences were generated, and with each library averaged 5.1 Gb. Conclusions: These important data might improve our understanding of reproductive biology of Luhua chicken by providing comprehensive gene expression information at transcriptional level. We indicate that our approach will contribute to the isolation of effective molecular markers that can be used in genetic breeding programs in Chinese domestic Luhua chicken.

Project description:Overfeeding reduces laying performance in broiler breeder hens (Gallus gallus domesticus). To unravel the effect of feeding regimes on energy metabolism and egg production, high-throughput RNA sequencing was utilized to identify differentially expressed genes (DEGs) in ovary, liver and adipose tissues of broiler chickens under ad libitum and restricted feeding. The transcriptome analysis showed that 289, 388 and 204 DEGs were identified in the adipose tissue, liver and ovary, respectively, between ad libitum and restricted feeding groups. Bioinformatics analysis by STRING further revealed DEGs were significantly enriched in phagosome pathway, lipid transport and location biological process, and the molecular function of lipid transporter activity and nutrient reservoir activity in ovary; the pathways of “steroid hormone biosynthesis” and “metabolism of xenobiotics by cytochrome P450”, and the molecular function of nutrient reservoir activity in adipose tissue; the metabolic pathways, Jak-STAT signaling pathway and PPAR signaling pathway in liver.
