Protein mass spectrum analysis of serum from CKD and NC mice


ABSTRACT: As for serum (CKD vs. NC =4 vs. 4), the protein was extracted by homogenate and SDT lysis. We utilized immunoaffinity depletion of serum high-abundance proteins. Next, we did the following steps: SDS-PAGE separation, Filter-aided sample preparation (FASP Digestion), TMT labeling, peptide fractionation with reversed-phase (RP) chromatography and mass spectrometry analysis. Mass Spectrometry analysis included Easy nLC, LC-MS/MS and data analysis. Differentially expressed proteins between two groups were determined using the t test (the threshold values were set as P < 0.05 and 1.5-fold change).
