DIA-based proteomics analysis of a CRISPR/Cas9-edited Class I α-mannosidase mutant during tomato fruit ripening
Ontology highlight
ABSTRACT: Class I α-mannosidases (MNSs) are N-glycan processing enzymes that specifically remove α-1,2-mannose residues from N-glycans attached to proteins. MNSs are generally categorized into three protein subgroups: ER-, Golgi-, and ER degradation-enhancing α-mannosidase (EDEM)-MNS. Although mutants of MNSs, which play roles in root development and cell wall biosynthesis, have been reported in Arabidopsis, high-fidelity datasets of the target N-glycoproteins represented by these MNS mutants have not yet been well established in plants. In this study, we focused on SlMNS1 as an N-glycogene and generated a CRISPR/Cas9-glycoengineered mutant to obtain stable loss-of-function slmns1 mutants. We then used DIA-based proteomics to analyze proteomic changes in the mutant compared to wild-type Ailsa Craig tomatoes at various fruit ripening stages: mature green, breaker, breaker plus 3 days, and breaker plus 7 days.
