Glioblastoma-derived extracellular vesicles facilitate transformation of astrocytes via reprogramming oncogenic metabolism

Project description:Background: Glioblastoma (GBM) may arise from brain astrocytes through a multistep process that occurs by the temporal accumulation of genetic mutations. Here, we explore whether GBM-derived extracellular vesicles (EVs) may facilitate neoplastic transformation and malignant growth of astrocytes. Methods: We utilized conditioned medium (CM) of glioma cell and stem cell, its sequential filtration, diverse cell-based assays, RNA sequencing, and metabolic assays to compare the effects of EV-containing and EV-depleted CM on the transformation of human and mouse astrocytes in vitro and tumor growth in vivo. Results: GBM EVs facilitated the neoplastic growth of astrocytes pre-transformed by oncogenic mutations or viruses but were unable to transform normal human and mouse astrocytes. GBM EVs induced proliferation, self-renewal, and colony formation of pre-transformed astrocytes, and enhanced astrocytoma growth in a mouse allograft model. Analysis of extracellular RNAs (exRNAs) in GBM identified multiple full-length mRNAs encoding ribosomal proteins, oxidative phosphorylation, and glycolytic factors. GBM EVs appear to reprogram astrocyte metabolism by inducing a shift in gene expression that may be partly associated with EV-mediated mRNA transfer from glioma cells. Conclusions: Our study suggests a novel EV/exRNA-mediated mechanism contributing to astrocyte transformation via metabolic reprograming and implicates horizontal mRNA transfer in this process.

Gene expression profiling of human gliomas and human glioblastoma cell lines

Project description:To identify signaling pathways that are differentially regulated in human gliomas, a microarray analysis on 30 brain tumor samples (12 primary glioblastomas (GBM), 3 secondary glioblastomas (GBM-2), 8 astrocytomas (Astro) and 7 oligodendrogliomas (Oligo)) and on 5 glioblastoma cell lines (LN018, LN215, LN229, LN319 and BS149) was performed. Normal brain tissue (NB) and normal human astrocytes (NHA) were used as a control. Kinase expression in each tumor was compared to expression in normal brain and expression values from normal human astrocytes were used as an additional control. Frozen tissue samples of human gliomas and normal brain obtained from the operating room were processed according to the guidelines of the Ethical Committee of the University Hospitals of Basel. Human brain tumor cell lines were derived from human patients. The BS149 cell line was generated at the University of Basel, Switzerland while the “LN” series were a kind gift of Erwin van Meir in Lausanne, Switzerland. Normal human astrocytes (NHAs) were purchased by Cambrex (Walkersville, MD) and cultured according to manufacturer’s recommendations.

Exosome-transmitted RVT1 from reactive astrocytes promotes recurrence glioblastoma proliferation

Project description:Glioblastoma (GBM) remains among the most lethal brain tumors despite improvements in treatment. Surgical resection is a universal component of glioma therapy. However, the mechanism underlying postoperative recurrence remains a challenge due to limited information regarding the changes that occur in the postoperative microenvironment. Here, we show that a subtype of proinflammatory reactive astrocytes designated is induced by inflammatory cytokines or injury. We show that reactive astrocytes can secrete the long noncoding RNA (lncRNA) LOC646762 to promotes recurrence glioblastoma proliferation in an exosome-dependent manner.We firstly named this reactive astrocyte associated lnRNA as “lnc-RVT1”.

Project description:Glioblastoma is a fatal and devastating primary brain tumour that poses significant challenges to accurate tumour monitoring and patient treatment. Substantial efforts are underway to develop accurate non-invasive liquid biopsy methods that can routinely assess glioblastoma markers in patient body fluids. We have recently shown that patient urine offers ready access to glioblastoma biomarkers within small extracellular vesicle populations (EVs; <200 nm membrane particles) and can accurately diagnose glioblastoma and reflect changes in glioblastoma pathophysiology. However, little is known about other urinary-EV populations in glioblastoma, and their capacity as biomarkers. In this study, we compared the biochemical properties of small (< 200 nm) and large (200-1000 nm) urinary-EVs using fourier-transform infrared spectroscopy and data independent acquisition mass spectrometry and identified significantly changing molecular distributions across the two EV populations in glioblastoma states. We also determined putative glioblastoma biomarkers associated with large urinary-EV biomarkers by ranking significant proteins with a logistic regression model. Our analysis revealed seven proteins capable of perfectly distinguishing all glioblastoma patients from healthy controls (AUC=1) and accurately classifying 96.7% of all cases as glioblastomas. Many significant large urinary-EV proteins also displayed consistent trends at different glioblastoma clinical timepoints, with protein abundance shifts post-surgery that return to pre-surgical levels at recurrence. Our findings demonstrate that both EV populations facilitate easy access to glioblastoma biomarkers for clinical diagnostics. Putative urinary-EV biomarkers described here merit further validation using comprehensive sets of longitudinal cohorts of glioblastoma urine.

A 5’ Sequence of Human HMGB2 Gene for Transcriptional Targeting of Glioblastoma

Project description:Achievement of specific tumor cell targeting remains a challenge for glioma gene therapy. We report here the identification and characterization of a 5’ sequence of human HMGB2 gene for transcriptional targeting to glioblastoma. We performed microarray analysis and found HMGB2 as one of the genes that had a low level of expression in normal human astrocytes, but was significantly up-regulated in glioblastoma cells. Real-time PCR quantification revealed increase in HMBG2 expression level in glioblastoma tissues and cells between 11 to 79 fold over that in normal human brain tissue. With progressive truncation of a 5’-upstream sequence of the HMGB2 gene, we identified a 500-bp fragment that displayed a high transcriptional activity in glioblastoma cells, but a low activity in normal brain cells. Using the sequence to drive the expression of the herpes simplex virus thymidine kinase gene in the context of a baculoviral vector, glioblastoma cells died in the presence of ganciclovir, whereas normal human astrocytes and neurons were not affected. We further confirmed that after intra-tumor injection, the baculoviral vector effectively suppressed the growth of human glioblastoma cells in a mouse xenograft model. Our results suggest that the 5’-upstream sequence of the HMGB2 gene can be used as an efficient, tumor-selective promoter in targeted vectors for glioblastoma gene therapy. U251 cells (n=3) genes level expression were compared to that of normal astrocytes (n=3) to find overexpressed genes in glioblastoma.
