Identification of RAC1 in Promoting Brain Metastasis of Lung Adenocarcinoma using Single-cell Transcriptome Sequencing

Project description:Brain metastases are common in lung adenocarcinoma (LUAD) patients, and by far, the metastasis mechanisms are not fully understood. We performed a comprehensive single-cell level transcriptomic analysis on one LUAD patient with CTC, primary tumor tissue and metastatic tumor tissue using scRNA-seq approach to identify metastasis related biomarkers. Further scRNA-seq were performed on 7 patients to validate the cancer metastatic hallmark. with single cells collected from either metastatic or primary LUAD tissues. we obtained a more comprehensive picture over lung cancer metastasis in the single-cell level, giving a new perspective to the role of RAC1 in the LUAD brain metastasis, and related pathways to participate in the metastasis process.

CBX7 Promotes Lung Adenocarcinoma Metastasis by Transcriptionally Repressing SOCS3

Project description:In lung adenocarcinoma (LUAD), metastasis is a leading cause of mortality and notably tends to occur at an early stage of the disease. CBX7, a component of canonical Polycomb Repressive Complex 1 (PRC1) which mediates transcriptional repression, exhibits complex and context-dependent functions across different types of cancer. However, the roles of CBX7 in LUAD metastasis remain largely uncharacterized. Here we report that overexpression of CBX7 promotes, whereas its knockdown suppresses, LUAD cell migration and invasion in vitro. In vivo studies further confirm the pro-metastatic role of CBX7 in LUAD. Mechanistically, CBX7 suppresses the expression of SOCS3, which has been reported to be involved in cancer cell migration and invasion. At the molecular level, CBX7 binds to SOCS3 transcription start site (TSS) downstream region to establish monoubiquitination of histone H2A at lysine 119 (H2AK119ub), leading to transcriptional repression of SOCS3. Furthermore, knockdown of SOCS3 rescues the reduced migration and invasion caused by CBX7 depletion in LUAD cells. Together, our findings identify CBX7 as a positive regulator of LUAD metastasis and suggest its potential as a therapeutic target for LUAD treatment.

Overexpressed Malat1 drives metastasis through inflammatory reprogramming of lung adenocarcinoma microenvironment

Project description:Although the expression of some long noncoding RNAs (lncRNAs), including MALAT1 (metastasis-associated lung adenocarcinoma transcript 1), is predictive of metastasis, their impact and mechanism of action remain elusive. Here we use CRISPR activation (CRISPRa) to model MALAT1/Malat1 overexpression in patient-derived lung adenocarcinoma (LUAD) cell lines and in the autochthonous K-ras/p53 LUAD mouse model. The results indicate that Malat1 overexpression alone is sufficient to enable the progression of LUAD to metastatic disease. We show that overexpressed MALAT1/Malat1 enhances cell mobility and promotes the recruitment of pro-tumor macrophages to the tumor microenvironment through paracrine secretion of the CCL2/Ccl2 cytokine. We determine that Ccl2 upregulation results from an increase in global chromatin accessibility upon Malat1 overexpression. Importantly, macrophage depletion and Ccl2 blockade counteracted the effects of Malat1 overexpression. These data demonstrate that a single lncRNA can drive LUAD metastasis through reprogramming of the tumor microenvironment.

TGF-β induces an atypical EMT to evade immune mechanosurveillance in lung adenocarcinoma dormant metastasis

Project description:The heterogeneity of epithelial-to-mesenchymal transition (EMT) programs is manifest in the diverse EMT-like phenotypes occurring during tumor progression. However, little is known about the mechanistic basis and functional role of specific forms of EMT in cancer. Here we address this question in lung adenocarcinoma (LUAD) cells that enter a dormancy period in response to TGF-β upon disseminating to distant sites. LUAD cells with the capacity to enter dormancy are characterized by expression of SOX2 and NKX2-1 primitive progenitor markers. In these cells, TGF-β induces growth inhibition accompanied by a full EMT response that subsequently transitions into an atypical mesenchymal state of round morphology and lacking actin stress fibers. TGF-β induces this transition by driving the expression of the actin-depolymerizing factor gelsolin, which changes a migratory, stress fiber-rich mesenchymal phenotype into a cortical actin-rich, spheroidal state. This transition lowers the biomechanical stiffness of metastatic progenitors, protecting them from killing by mechanosensitive cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. Inhibiting this actin depolymerization process clears tissues of dormant metastatic cells. Thus, LUAD primitive progenitors undergo an atypical EMT as part of a strategy to evade immune-mediated elimination during dormancy. Our results provide a mechanistic basis and functional role of this atypical EMT response of LUAD metastatic progenitors and further illuminate the role of TGF-β as a crucial driver of immune evasive metastatic dormancy.

Transcriptomic profiling of Lung adenocarcinoma brain metastasis

Project description:Brain metastasis developed in nearly 40% of lung adenocarcinoma (LUAD) patients diagnosed with distant metastasis. There is lack of transcriptomic data of brain lesions from human lung adenocarcinoma patients. As part of the project to understanding the tumor microenvironment in brain metastasis of LUAD patients, we performed bulk RNA analysis on brain metastases from 6 LUAD patients. In order to understand the tumor intrinsic factors that potential shape the tumor microenvironment, we compared these data with bulk RNA sequencing data from 14 early stage and 11 late stage primary LUAD tumor from TCGA database. Pathway expression analysis showed a downregulation of pro-inflammatory signals in brain metastasis and upregulation of DNA synthesis and oxidative phosphorylation pathways related to rapid proliferation in brain lesions.

Transcriptional dissection by single-cell RNASeq analysis of the metastasis settings from a highly metastatic Patient-Derived Xenograft model of pancreatic ductal adenocarcinoma

Project description:Single cells were isolated from the main scenarios during tumor dissemination of a highly metastatic Patient-Derived Xenograft model (Panc-265) in order to discover potential therapeutic targets against the metastatic settings of PDAC. Primary tumor (n=37), liver metastasis (n=23) cells and circulating tumor cells (CTCs, n=10, in bloodstream) were subjected of single-cell RNAseq. Single-cell RNA sequencing analysis revealed that CTCs clustered separately from their matched primary and metastatic tumors and were characterized by low expression of ECM associated genes, high expression of cell cycle genes, a metabolic switch to oxidative phosphorylation and high expression of genes involved in ribosome biogenesis.

Transcriptomic hallmarks of tumor plasticity and stromal interactions in brain metastasis [BrainMet_SQ_2D]

Project description:We find that malignant LUAD tumor cells grown in the brain microenvironment display significantly altered transcriptomic profiles when compared to tumor cells grown in monolayer or subcutaneously. Additionally, we characterize the reciprocal neuroinflammatory response of the surrounding brain stroma to metastatic LUAD. This dataset can be utilized to explore the mechanisms of LUAD cell plasticity as well as the co-adaptation of the brain microenvironment to metastatic disease.
