Transcriptomic and proteomic analysis revealed the inhibition of carvacrol against garlic brassica

ABSTRACT: In order to prevent and control the specific pathogenic fungus of Fusarium argentifolia isolated from garlic brassica, the mechanism of action of carvacrol on Fusarium argentifolia was explored based on two levels of transcription and protein, so as to inhibit Fusarium argentifolia brassica.

Project description:Fusarium head blight (FHB) is a major disease of cereal crops caused by the fungus Fusarium graminearum (Fg). FHB affects the flowering heads (or spikes) and developing seeds. This study compare the gene expression profile in wheat spikelets (spk 2) inoculated with either water (mock treatment) or a pathogenic strain of Fusarium graminearum (WT); spikelets 2 were inoculated 24 hrs after a neighbour spikelet (spk 0) was treated with either water or F. graminerum mutant strain Tri6Δ or NoxABΔ. Spikelets 2 were sampled 8 and 24 hrs after the second treatment.

Tracing the transcriptomic changes of synthetic trigenomic allohexaploid in Brassica by RNA-Seq approach

Project description:We explored the transcriptomic changes of synthetic Brassica allohexaploid by comparing to its parents using a high-throughput RNA-Seq method. A total of 35644409 sequence reads were generated, and 32642 genes were aligned from the data. There were 29260, 29060 and 29697 genes identified in Brassica rapa, Brassica carinata, and Brassica allohexaploid, respectively. We screened differentially expressed genes (DEGs) by a standard of two-fold or greater change in expression and false discovery rate (FDR) no more than 0.001. As a result, 7397 DEGs were detected between Brassica hexaploid and its parents. A large proportion of the 3184 DEGs between Brassica hexaploid and its paternal parent B. rapa was involved in biosynthesis of secondary metabolites, plant-pathogen interaction, photosynthesis, and circadian rhythm. Between Brassica hexaploid and its maternal parent B. carinata, 2233 DEGs were screened. A lot of them had functions of plant-pathogen interaction, plant hormone signal transduction, ribosome, limonene and pinene degradation, photosynthesis, and also biosynthesis of secondary metabolites. Project description:Fusarium graminearum (teleomorph Gibberella zeae) is a prominent pathogen that infects major cereal crops, such as wheat, barley, and maize. To dissect cellular responses toward heat stress in the plant pathogenic fungus F. graminearum, we compared transcriptomes of the fungal cultures incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min. 6 samples examined: 24 h-old mycelia from complete medium (CM) of Fusarium graminearum wild-type Z-3639 were incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min.
