Proteins raw data using CT-1 in MDA-MB-231

ABSTRACT: The MDA-MB-231 cells were treated with CT-1 for 24 h. Then the cells were collected for proteomic analysis. SDT buffer was used for sample lysis and protein extraction. Protein digestion by trypsin was performed according to filter-aided sample preparation (FASP) procedure described by Matthias Mann. Then 100 μg peptide mixture of each sample was labeled using TMT reagent according to the manufacturer’s instructions. LC-MS/MS analysis was performed on a Q Exactive mass spectrometer that was coupled to Easy nLC for 60/90 min. The MS raw data for each sample were searched using the MASCOT engine embedded into Proteome Discoverer 1.4 software for identification and quantitation analysis.

Tumor-associated neutrophils (TANs) play a critical role in the progression and prognosis of triple-negative breast cancer (TNBC), with N2-type TANs known for their pro-tumor characteristics. This study introduces CT-1, a derivative of cryptotanshinone that effectively suppresses TNBC growth while selectively reducing the proportion of N2-type TANs within tumor tissue. Notably, CT-1 induces simultaneous ferroptosis in both N2-type TANs and TNBC cells, a dual mechanism that enhances its therapeut  ...[more]
