Project description:Lipid droplets (LDs) are dynamic organelles essential for lipid storage and organismal survival. Studies have highlighted the importance of glial function in brain LD formation during aging, however, the genes and mechanisms involved remain elusive. Here, we found that Ugt35b, a member of the UDP-glycosyltransferases that catalyze the transfer of glycosyl groups to acceptors, is highly expressed in glia and crucial for Drosophila lifespan. By integrating multi-omics data, we demonstrated that glial Ugt35b plays key roles in regulating glycerolipid and glycerophospholipid metabolism in the brain. Notably, we found that Ugt35b and Lsd-2 are co-expressed in glia and confirmed their protein interaction in vivo. Knockdown of Ugt35b significantly reduces LD formation by downregulating Lsd-2 expression, while overexpression of Lsd-2 partially rescues the shortened lifespan in glial Ugt35b RNAi flies. Our findings reveal the crucial role of glial Ugt35b in regulating LD formation to maintain brain lipid homeostasis and support Drosophila lifespan.

Project description:Chloroplast group IIA introns derive from bacterial ribozymes. Their splicing likely requires Maturase K (MatK), which has been largely inaccessible to functional analyses being itself a chloroplast intron-encoded protein. Here we used co-immunoprecipitation coupled with proteomics to identify proteins bound to MatK in tobacco (Nicotiana tabacum) seedlings. We used homoplastomic tobacco lines modified to express NtMatK-HA (line NtMatK C+) or control lines carrying only the selection marker (line NtMatK C-). Proteins co-precipitating with NtMatK-HA were purified via anti-HA beads and identified via shotgun proteomics (n=3 seedling pools). The mass spectrometry data were further processed using Philosopher followed by label-free quantification based on precursor intensities using IonQuant and statistical analysis using Amica.

Project description:Drosophila miRNAs show distinct change in isoform distribution pattern with age. Some miRNAs show accumulation of the short isoforms, while other miRNAs show the accumulation of the long isoforms with age. The increase of the long isoforms of some miRNAs reflects increased 2'-O-methylated miRNA isoforms with age. The increase in 2'-O-methylated miRNA isoforms reflected increased Ago2-loading, but not Ago1-loading of specific miRNA isoforms with age. This raised a question on whether there is global shift in small RNA loading pattern between Ago1 and Ago2 with age. To investigate change in small RNA loading pattern between Ago1 and Ago2 with age, we performed small RNA deep-sequencing of Ago1 vs Ago2-IP small RNAs at 3d and 30d in Drosophila. This analysis revealed global increase of miRNA loading into Ago2, but not into Ago1 with age. 3d and 30d FLAG-HA-Ago2 male flies were collected. Ago1 and Ago2 were immunoprecipitated by anti-Ago1 and anti-FLAG M2 beads respectively. RNA was purified from the beads, P32-labeled, and small RNA fraction was gel-purififed. Small RNA libraries were prepared using Illumina's TruSeq small RNA sample preparation kit (#RS-200-0012, Illumina, Inc. San Diego, CA), following the manufacturer's protocol. The libraries were multiplexed and sequenced on HiSeq2000 platform (Illumina).

Regenerated nerve tissues from GDNF-Gel/HA-Mg group and negative control group were collected for RNA-seq, aiming to explore the potential molecular mechanism of GDNF-Gel/HA-Mg in peripheral nerve regeneration.

Project description:Much focus has shifted towards understanding how glial dysfunction contributes to age-related neurodegeneration due to the crucial roles glial cells play in maintaining healthy brain function. Cell-cell interactions, which are largely mediated by cell-surface proteins, control many critical aspects of development and physiology; as such, dysregulation of glial cell-surface proteins in particular is hypothesized to play an important role in age-related neurodegeneration. However, it remains technically difficult to profile glial cell-surface proteins in intact brains. Here, we applied a cell-surface proteomic profiling method to glial cells from intact brains in Drosophila. Importantly, this enabled us to fully profile cell-surface proteomes in-situ, preserving native cell-cell interactions that would be omitted using more traditional proteomics methods. Applying this platform to young and old flies, we investigated how glial cell-surface proteomes change during aging. We identified candidate genes predicted to be involved in normal brain aging, including several associated with neural development and synapse wiring molecules not previously thought to be particularly active in glia. Through a functional genetic screen, we identified one surface protein, DIP-B, which is down-regulated in old flies and can increase fly lifespan when overexpressed in adult glial cells. We further performed whole-head single-nucleus RNA-seq, and revealed that DIP-B overexpression mainly impacts glial and fat cells. We also found that glial DIP-B overexpression was associated with improved cell-cell communication, which may contribute to the observed lifespan extension. Our study is the first to apply in-situ cell-surface proteomics to glial cells in Drosophila, and to identify DIP-B as a potential glial regulator of brain aging.

Project description:First, the snRNP component U1A was HA-tagged and the intron of the RabX13 gene was MS2-tagged and amoeba transformats were established with such constructs. Next, amoeba transformants were UV cross-linked and their nucler extracts were immunoprecipitated (IP) with anti-HA agarose or MS2-spharose beads. Precipitates were subjected to tandem mass spectrometry (MS/MS) analyses. MS2-IP helped to discriminate the nuclear roles (chromatin-, co-transcriptional-, splicing-related), of the proteins probed.

Project description:<p>Viral studies of Drosophila melanogaster typically involve virus injection with a small needle, causing post-injury a wounding/wound healing response, in addition to the effects of viral infection. However, the metabolic response to the needle injury is understudied, and many viral investigations neglect potential effects of this response. Furthermore, the wMel strain of the endosymbiont bacterium Wolbachia pipientis provides anti-viral protection in Drosophila. Here we used NMR-based metabolomics to characterise the acute wounding response in Drosophila and the relationship between wound healing and the Wolbachia strain wMel. The most notable response to wounding was found on the initial day of injury and lessened with time in both uninfected and Wolbachia infected flies. Metabolic changes in injured flies revealed evidence of inflammation, Warburg-like metabolism and the melanisation immune response as a response to wounding. In addition, at five days post injury Wolbachia infected injured flies were metabolically more similar to the uninjured flies than uninfected injured flies were at the same time point, indicating a positive interaction between Wolbachia infection and wound healing. This study is the first metabolomic characterisation of the wound response in Drosophila and its findings are crucial to the metabolic interpretation of viral experiments in Drosophila in both past and future studies.</p>
