Project description:In this work, we detect the effect of EVs derived from M. bovis-infected BMEC cells (M. bovis NX2-EVs) on regulating the inflammatory response of BoMac. The results indicate M. bovis-infected BMEC cells promote the inflammatory response in BoMac cells by releasing EVs. We furtherly identify the miRNAs in EVs involved in promoting inflammatory response. Understanding their cargo content will provide new insights into the molecular mechanisms of these respective EVs.

Effect of M . bovis NX2-EVs on gene expression in BoMac cells

Project description:Our research shows that EVs derived from M. bovis-infected BMEC cells (EVs-M. bovis NX2) could regulate BoMac cell injury by inducing inflammation. To detect the effect of EVs-M. bovis NX2 on gene expression in BoMac cells, we incubated Ctrl-EVs and EVs-M. bovis NX2 with BoMac cells for 12h, 24h. Our data provide globaly change of mRNA profile caused by EVs-M. bovis NX2.

Differentially enriched miRNAs in EVs derived from young and aged osteocytes of mice

Project description:Vascular calcification often occurs with osteoporosis, a contradictory association called “calcification paradox”. We find that extracellular vesicles (EVs) released from aged bone matrix (AB-EVs) during bone resorption favor adipogenesis rather than osteogenesis of BMSCs and augment calcification of vascular smooth muscle cells (VSMCs). Intravenous or intramedullary injection of AB-EVs promotes bone-fat imbalance and exacerbates Vitamin D3 (VD3)-induced vascular calcification in young or old mice. To explore the involvement of miRNAs in the AB-EVs-induced promotion of adipocyte formation and vascular calcification, the Agilent miRNA array was conducted to compare the miRNA expression profiles in AB-EVs and YB-EVs from mouse bone specimens. Our study uncovers the role of AB-EVs as a messenger for calcification paradox by transferring functional miRNAs.

Project description:Microvesicles (MVs) are a subtype of extracellular vesicle (EVs) released by cells, which play important roles in cell-cell communication. The role of host cell-derived MVs in host-pathogen interactions remains to be defined in the context of mycobacterial infection. In this study, we characterized MVs that were isolated from mouse RAW 264.7 cells either uninfected or infected with Mycobacterium abscessus (M.ab), one of the most common non-tuberculous mycobacterial (NTM) pathogens in humans, in cell culture. The proteomic profile of purified MVs was determined by mass spectrometry (MS)-based proteomics.

Neutrophils infected with Leishmania donovani

Project description:In the context of studying visceral leishmaniasis, neutrophils infected with Leishmania donovani have been compared to uninfected neutrophils. Compared time points are 0, 6 and 24 hours post infection. Neutrophils of three human donors have been used. Overall 6 samples for infected neutrophils at time point 6 hours and 6 samples for infected neutrophils at time point 24 hours exist, including three biological samples and two technical samples. Uninfected neutrophils represent 3 samples at time point 0 hours, 3 samples at time point 6 hours and 3 samples at time point 24 hours. Transcriptome of Leishmania donovani culture has been assessed in two replicates.
