Project description:Despite their distinct biology, granulosa cell tumours (GCTs) are treated the same as other ovarian tumours. Intriguingly, a recurring somatic mutation in the transcription factor Forkhead Box L2 (FOXL2) 402C>G has been found in nearly all GCTs examined. This investigation aims to identify the pathogenicity of mutant FOXL2 by studying its altered transcriptional targets. The expression of mutant FOXL2 was reduced in the GCT cell line KGN, and wildtype and mutant FOXL2 were overexpressed in the GCT cell line COV434. Comparisons were made between the transcriptomes of control cells and cells altered by FOXL2 knockdown and overexpression, to detect potential transcriptional targets of mutant FOXL2. Plasma proteomics to discover diagnostic biomarker for Alzheimer's disease

Project description:The project aims to find diagnostic biomarkers for Alzheimer's disease in plasma. We applied discovery based proteomics was used to detect changes in proteins due to Alzheimer's disease using plasma samples collected from four study groups (African American Alzheimer's disease & cognitively normal, non-Hispanic-White Alzheimer's disease & cognitively normal). The data emphesizes the need for inclusion in Alzheimer's disease research.

Project description:This study investigates the effect of NCAPD2 knockdown on lung adenocarcinoma (LUAD) by performing RNA sequencing on PC9 cell lines. The experiment included three control PC9 cells and three PC9 cells in which NCAPD2 was knocked down using shRNA. This RNA sequencing experiment aims to elucidate the role of NCAPD2 in regulating gene expression associated with tumor proliferation, migration, and survival in LUAD. Comparative Proteomic Profiling of Exosomes Derived from Sema6A-Knockdown hPMVECs

Project description:This project aims to characterize the proteomic landscape of extracellular vesicles (EVs) released by human pulmonary microvascular endothelial cells (hPMVECs) following Sema6A gene knockdown. Using lentiviral-mediated RNA interference, we generated stable Sema6A-knockdown cell lines (*sh-Sema6A*) alongside negative control cells (sh-NC). Exosomes were isolated from conditioned media of both groups via ultracentrifugation and validated for EV-specific markers (e.g., CD63, TSG101). Subsequent high-resolution tandem mass spectrometry (LC-MS/MS)-based proteomic analysis will quantitatively compare protein cargoes between *sh-Sema6A*-EVs and sh-NC-EVs. This study seeks to identify Sema6A-regulated exosomal proteins implicated in endothelial cell communication, angiogenesis, or vascular pathophysiology, providing mechanistic insights into semaphorin signaling pathways in the pulmonary vasculature.
