Maresin 1 regulates inflammation in gingival mesenchymal stem cells under LPS-induced inflammatory states

ABSTRACT: Periodontitis is a chronic inflammatory condition marked by an imbalance between proinflammatory and proresolution mediators, which affects the tissues supporting the teeth. Maresin1 (MaR1) is a specialized pro-resolving lipid mediator (SPM) produced by macrophages, known for its role in regulating inflammatory processes. This study aimed to investigate the effects of MaR1 at varying concentrations on human gingival mesenchymal stem cells (hGMSCs) within inflammatory environments.

Comparative Proteomics of Tissue and Gingival Crevicular Fluid in Periodontitis

Project description:Periodontitis is an inflammatory condition affecting periodontal tissues, such as the gingiva and alveolar bone, surrounding the teeth. However, previous research has focused on analyzing a single sample type, such as tissue, blood, or gingival crevicular fluid (GCF), which may limit the comprehensive understanding of the complex pathology of periodontitis. We conducted a proteomic analysis using liquid chromatography-mass spectrometry (LC-MS) on the periodontal tissues and GCF from patients with periodontitis. This analysis distinguished distinct proteomic signatures between the two sample types, with 3,803 proteins being identified in periodontal tissue and 910 proteins in GCF, underscoring the significant variances pertinent to periodontal health. Proteins such as S100A12, MPO, APOA1, CTSG, and PSMA1 were markedly upregulated in GCF, suggesting their potential as key markers of neutrophil activity and systemic inflammation. These findings provide profound insights into both the local and systemic pathophysiological processes underlying periodontitis, emphasizing the utility of GCF as a non-invasive medium for reflecting comprehensive disease states.

Single-cell transcriptomic atlas of mouse gingival stroma in homeostasis and inflammation

Project description:The onset of periodontitis involves the response of gingival stroma to inflammatory stimulus. The cell constitution of gingival stroma, however, has never been mapped at the resolution of single cell. In this study, we used single-cell mRNA sequencing to establish cell atlas in mouse gingival stroma. Epithelial, mesenchymal, pericyte and neuronal clusters were successfully identified and their subpopulation and response to ligature-induced inflammation were also examined. While shifts in epithelial cell gene profile and proportion recapitulated the epithelium breakdown during periodontitis, mesenchymal cluster maintained its proportion and gene expression patterns.

MicroRNAs and their target genes in gingival tissues.

Project description:To gain insights into the in vivo function of miRNAs in the context of periodontitis, we examined the occurrence of miRNAs in healthy and diseased gingival tissues and validated their in silico-predicted targets through mRNA profiling using whole-genome microarrays in the same specimens. Eighty-six individuals with periodontitis contributed 198 gingival papillae: 158 'diseased' (bleeding-on-probing, PD > 4 mm, and AL M-bM-^IM-% 3 mm) and 40 'healthy' (no bleeding, PD M-bM-^IM-$ 4 mm, and AL M-bM-^IM-$ 2 mm). Expression of 1,205 miRNAs was assessed by microarrays, followed by selected confirmation by quantitative RT-PCR. Predicted miRNA targets were identified and tested for enrichment by Gene Set Enrichment Analysis (GSEA). Enriched gene sets were grouped in functional categories by DAVID and Ingenuity Pathway Analysis. One hundred fifty-nine miRNAs were significantly differentially expressed between healthy and diseased gingiva. Four miRNAs (hsa-miR-451, hsa-miR-223, hsa-miR-486-5p, hsa-miR-3917) were significantly overexpressed, and 7 (hsa-miR-1246, hsa-miR-1260, hsa-miR-141, hsa-miR-1260b, hsa-miR-203, hsa-miR-210, hsa-miR-205*) were underexpressed by > 2-fold in diseased vs. healthy gingiva. GSEA and additional filtering identified 60 enriched miRNA gene sets with target genes involved in immune/inflammatory responses and tissue homeostasis. This is the first study that concurrently examined miRNA and mRNA expression in gingival tissues and will inform mechanistic experimentation to dissect the role of miRNAs in periodontal tissue homeostasis and pathology. The dataset comprise 200 gingival tissue samples from 86 patients with periodontitis. All samples are included in this series, but 2 where not included in the study, as they did not pass our QC. Each patient has one or more healthy and disease sample and samples can be of chronic or aggressive periodontitis type.

Altered gene expressions of leukocyte transendothelial migration and cell communication pathways in periodontitis-affected gingival tissues

Project description:Gene expressions relate to the pathogenesis of periodontitis and have a crucial role in local tissue destruction and susceptibility to the disease. The aims of the present study were to explore comprehensive gene expressions/transcriptomes in periodontitis-affected gingival tissues, and to identify specific biological processes. The purpose of the present study was 1) to compare comprehensive gene expression/transcriptomes of periodontitis-affected gingival tissues with those of healthy tissues by using microarray and data mining technologies, and 2) to analyze significantly differentially expressed genes which belong to pathological pathways in periodontitis by qRT-PCR. Two distinct gingival samples including healthy and periodontal-affected gingiva were taken from 3 patients with severe chronic periodontitis. Total RNAs from 6 gingival tissue samples were used for microarray and data-mining analyses. Comparisons, gene ontology, and pathway frequency analyses were performed and identified significant biological pathways in periodontitis. Quantitative reverse transcription real-time polymerase chain reaction (qRT-PCR) analyse using 14 chronic periodontitis patients including 3 patients listed above and 14 healthy individuals showed 9 differentially expressed genes in leukocyte migration and cell communication pathways.

Transcriptomes in Healthy and Diseased Gingival Tissues

Analysis of the gingival tissue transcriptome in states of periodontal health and disease may reveal novel insights of the pathobiology of periodontitis. Keywords: gingival tissue disease state analysis Ninety non-smokers, 63 with chronic and 27 with aggressive periodontitis, each contributing with >=2 “diseased” interproximal papillae (with bleeding on probing, probing pocket depth >=4mm, and clinical attachment loss >=3mm) and a “healthy” papilla, if available (no BoP, PPD<=4mm and CAL<=2mm). RNA was extracted, amplified, reverse transcribed, labeled, and hybridized with AffymetrixU133Plus2.0 arrays. Transcriptome analysis was conducted on a total of 247 samples (from 183 diseased and 64 healthy sites).

Transcription profiling of gingival tissue from patients with periodontitis: Bacterial Correlates of Gingival Gene Expression

Project description:We investigated the association between subgingival bacterial profiles and gene expression patterns in gingival tissues of patients with periodontitis. A total of 120 patients undergoing periodontal surgery contributed with a minimum of two interproximal gingival papillae (range 2-4) from a maxillary posterior. Prior to tissue harvesting, subgingival plaque samples were collected from the mesial and distal aspects of each papilla. Gingival tissue RNA was extracted, reverse-transcribed, labeled, and hybridized with whole-genome microarrays (310 in total)
