Project description:Tandem Mass Tag (TMT)-based quantitative proteomic analysis of tomato soil borne pathogen Fusarium oxysporum f. sp. radicis-lycopersici growth, and metabolism when treated with plant natural volatile organic compounds linalool. The Forl strain was cultured on PDA supplied with 0.8 mL/L linalool for 6 days at 25°C. The fungal strain on PDA supplied with only 0.1% Tween80 was cultured as the control. Three biological replicates were established for each treatment.

Ear rot in maize is a destructive disease globally caused by Fusarium moniliforme , due to decrease of grain yield and increase of risks in raising livestock by mycotoxins production. Plants have developed various defense pathways to cope with pathogens. We used microarrays to detail the global programme of gene expression during the infection process of Fusarium moniliforme in its host plant to get insights into the defense programs and the host processes potentially involved in plant defense against this pathogen.

Project description:Facultative heterochromatin marked by histone H3 lysine 27 trimethylation (H3K27me3) is an important regulatory layer for secondary metabolite (SM) gene silencing and important for fungal development in the genus Fusarium. While this histone mark is essential in some (e.g., the rice pathogen Fusarium fujikuroi), it appears dispensable in other fusaria. Here, we show that deletion of FpKMT6 is detrimental but not lethal in the plant pathogen Fusarium proliferatum, a member of the Fusarium fujikuroi species complex (FFSC). Loss of FpKmt6 results in aberrant growth, and expression of a large set of previously H3K27me3-silenced genes is accompanied by increased H3K27 acetylation (H3K27ac) and an altered H3K36me3 pattern. Next, H3K9me3 patterns are affected in ∆fpkmt6, indicating a crosstalk between both heterochromatic marks that became even more obvious in a strain deleted for FpKMT1 encoding the H3K9-specific histone methyltransferase. In ∆fpkmt1, all H3K9me3 marks present in the wild-type strain are replaced by H3K27me3, a finding that likely explains the subtle phenotype of ∆fpkmt1 strains which stands in marked contrast to other filamentous fungi. A large proportion of SM-encoding genes is allocated with H3K27me3 in the wild-type strain, and loss of H3K27me3 result in elevated expression of 49% of them. Interestingly, genes involved in the biosynthesis of the phytohormones gibberellins (GA) are among the most upregulated genes in ∆fpkmt6. Although several FFSC members harbor GA biosynthetic genes, its production in planta is largely restricted to F. fujikuroi with few exceptions. We show that GA gene silencing is mediated by H3K27me3 in F.proliferatum and in at least one additional FFSC member, possibly outlining the distinct lifestyles of these notorious plant pathogens.

Project description:Fungal effectors are key determinants of disease development, promoting pathogen growth and spreading in plant tissues. In the wheat/Fusarium graminearum (Fg) interaction determining the Fusarium Head Blight disease (FHB), the molecular bases of Fg aggressiveness remain widely unknown. The aim of this work is to provide a better understanding of Fusarium graminearum aggressiveness determinism during its interaction with bread wheat. Phenotyping in controlled conditions of three fungal strains was carried out on three wheat cultivars contrasted by their susceptibility to FHB. Qualitative and quantitative characterizations of the three strain's whole proteomes expressed in planta have been performed with special interest on effectors and proteins coded by Fg aggressiveness-related genes. Considering all fusaried wheat samples, this experimentation identified 615 unique Fg proteins, including 583 identified in all samples. Furthermore, 71 putative Fg effectors were found in the identified proteins. Among all the fungal proteins quantified, nearly 76% showed significant abundance differences between the Fg strains. Two-way ANOVA were computed to evaluate the contribution of the "Strain" and "Cultivar" factors on the Fg protein abundances. Abundances that were deemed significant to each factor enabled a categorization of the proteins according to the factor(s) that drive(s) the abundance differences. This included: (i) Fg proteins whose abundance differences were specifically explained by the genetic background of the strain (Strain factor, Strain_effect proteins), (ii) Fg proteins whose abundance differences were only explained by the genetic background of the host plant (Cultivar factor, Cultivar_effect proteins), (iii) Fg proteins whose abundance differences were explained by both factors (Strain+Cultivar proteins), and (iv) Fg proteins whose abundance differences observed between strains were dependent on the host cultivar (Strain×Cultivar proteins). Considering each factor individually, a total of 139 Strain_effect proteins, 82 Cultivar_effect proteins, 117 Strain+Cultivar proteins and 2 Strain×Cultivar proteins were identified.

This study compare the gene expression profile in wheat spikelets (spk 2) inoculated with either water (mock treatment) or a pathogenic strain of Fusarium graminearum (WT); spikelets 2 were inoculated 24 hrs after a neighbour spikelet (spk 0) was treated with either water or F. graminerum mutant strain Tri6Δ or NoxABΔ. Spikelets 2 were sampled 8 and 24 hrs after the second treatment.

Project description:The plant pathogenic fungus Fusarium graminearum (Fgr) creates economic and health risks in cereals agriculture. Fgr causes head blight (or scab) of wheat and stalk rot of corn, reducing yield, degrading grain quality and polluting downstream food products with mycotoxins. Fungal plant pathogens must secrete proteases to access nutrition and to breakdown the structural protein component of the plant cell wall. Research into the proteolytic activity of Fgr is hindered by the complex nature of the suite of proteases secreted. We used a systems biology approach comprising genome analysis, transcriptomics and label-free quantitative proteomics to characterise the peptidases deployed by Fgr during growth. A combined analysis of published microarray transcriptome datasets revealed seven transcriptional groupings of peptidases based on in vitro growth, in planta growth, and sporulation behaviours. An orbitrap MS/MS proteomics technique defined the extracellular proteases secreted by Fusarium graminearum.

Expression analyses performed in the soil-borne plant pathogen Fusarium oxysporum revealed that almost all copper starvation-induced genes are downregulated in the absence of the regulator Mac1. Project description:The colonization of Capsicum annuum roots by Fusarium oxysporum Fo47 induces resistance responses on the plant. Fo47 is a non-pathogenic strain of Fusarium oxysporum. Fo47 colonizes only the most outer layers of the root surface but it does not colonize inner tissues. Pre-treatment of roots with Fo47 reduces the symptom development produced by later pathogen inoculation. The expression of genes in distal tissues was determined by microarray analysis of stems of Fo47-treated plants. Capsicum annuum samples were analyzed using Affymetrix chips of the close-related species Solanum lycopersicum.
