Sequencing and bioinformatics analysis of exosome-derived miRNAs in mouse models of pancreatic injury induced by OSA

Project description:The role of exosomal miRNAs involved in the pathogenesis and development of pancreatic injury caused by obstructive sleep apnea (OSA) remains unclear. Here, miRNA sequencing (miRNA-seq) was used to investigate the expression profile of exosomal miRNAs in pancreatic tissue in a mouse model of chronic intermittent hypoxia (CIH) -induced pancreatic injury. A total of 55 newly predicted miRNAs and 277 existing miRNAs were identified in pancreatic tissue exosomes from the mouse model of chronic intermittent hypoxic-induced pancreatic injury. 3 up-regulated and 33 down-regulated miRNAs with differential expression were identified. 8 exosome-derived miRNAs were selected to construct CNC networks to predict target genes. Some targets and pathways were elucidated by GO analysis and KEGG analysis. Our work first comprehensively studied the expression profile of exosome-derived miRNA in pancreatic tissue of mouse models of OSA-induced pancreatic injury, bringing fresh perspectives on managing pancreatic injury brought on by OSA.

Project description:Changes in the secretion profile of visceral-pancreatic white adipose tissue (pWAT) due to diet-induced obesity are partially responsible for increased beta cell replication, suggesting that a crosstalk between pWAT and beta cells may play a role in regulating beta cell plasticity. The molecular mechanisms underlying this cross-talk are still not fully understood. The aim of this study was to integrate transcriptomic, proteomic and metabolomic data to unravel the cross-talk between adipose tissue and pancreatic islets during evolution of obesity. Pancreatic islets from control lean and cafeteria diet fed obese rats were obtained. RNA was extracted and processed for further hybridization on Affymetrix microarrays (GeneChip Rat Genome 230 2.0 (Affymetrix, Santa Clara, CA)).

CagriSema drives weight loss in rats by reducing energy intake and preserving energy expenditure

Project description:CagriSema is a combination of amylin (cagrilintide) and glucagon-like peptide-1 (semaglutide) analogues being developed for weight management. We quantified CagriSema’s action on energy intake and expenditure in rats. CagriSema caused 12% weight loss and a 39% reduction in food intake. In contrast, pair-feeding caused less weight loss while weight-matching required a 51% decrease in food intake. Approximately 1/3 of CagriSema’s weight loss efficacy was from an energy expenditure effect, blunting of metabolic adaptation, which is likely a hallmark of successful treatment of obesity.

Extracellular vesicles from obese visceral adipose promote pancreatic cancer development and resistance to immune checkpoint blockade therapy.

Project description:Obesity is correlated with multitype of cancer development, and pancreatic ductal adenocarcinoma (PDAC) in obese patients shows dismal prognosis and resistance to immune checkpoint blockade (ICB) therapy. The molecular mechanism is largely unknown. Here we show that obese visceral adipose tissues (VAT) can communicate with distant PDAC by delivering extracellular vesicle (EV) carrying signal molecules. We reveal that PDAC cells can take in VAT-EVs to their lysosomes where EVs-delivered Cathepsin A (Ctsa) stabilize ribonuclease Rnaset2b to produce free pseudouridines from RNA cleavage. Pseudouridines further activate mast cells via mediating an increase in reactive oxygen species (ROS) generation, which leads to a decrease in the distribution of H3K27me3 at the gene promoter, ultimately inhibiting CD8+ T cell activity, forming an immunosuppressive tumor microenvironment which enhances cancer progression and ICB resistance. We also demonstrate harmful effects of VAT-EV CTSA-pseudouridine-mast cell axis for obesity-related PDAC. Animal experiments indicate that Ctsa knockdown effectively enhances ICB efficacy on PDAC. Our study uncovers a mechanism connecting obesity and cancer which holds promise for developing new therapeutic strategy for obesity-related cancers.
