Project description:HEK293T cells are a widely used experimental model in cell biology. However, the presence and robustness of intrinsic circadian rhythmicity in HEK293T cells remain controversial. To address this issue, we synchronized HEK293T cells with dexamethasone and initiated sample collection 24 hours after synchronization. Cells were harvested every 4 hours over a 48-hour period, followed by transcriptomic (RNA-seq) analysis.

Project description:To identify the major E3 ligase for c-Myc ubiquitylation in HEK293T cells, we overexpressed FLAG-tagged c-Myc in HEK293T cells and treated with MLN4924. and analyzed the interactors by immunoprecipitation and MS. Cell lysates were subjected to immunoprecipitation with antibodies to FLAG, and the immunoprecipitated proteins were analyzed by LC-MS/MS.
