The Role of Annexin A1 in Candida albicans and Candida auris infections in murine neutrophils

Project description:<p>Annexin A1 (AnxA1) is an anti-inflammatory protein present in different cells in biological systems, especially expressed in macrophages and neutrophils. Since neutrophils play an important role in infections and inflammatory processes and the AnxA1 and its relation in Candida spp. infections is not well understood, our study highlights a perspective in AnxA1 as a target protein in control the immune response during fungi infection. C57BL/6 wild-type (WT) and AnxA1 knockout (AnxA1-/-) peritoneal neutrophils were coinfected with Candida albicans or Candida auris for 4 hours. High levels of COX-2 were observed in WT and AnxA1-/- neutrophils infected with C. albicans and C. auris coinfection. In addition, AnxA1-/- neutrophils exhibited a marked increase of phosphorylated ERK, p-38 and JNK levels after coinfection with both Candida spp. Lipidomics approach shows that lack of AnxA1 produced a marked difference in lipid profile of control or coinfected neutrophil supernatants compared to WT, especially glycerophospholipids and glycerolipids. In summary, our results showed that endogenous AnxA1 regulates neutrophil response under fungal infection and its lipid membrane organization and metabolism.  </p>

Data in support of Proteomic analysis of synovial fluid from rheumatic arthritis and spondyloarthritis patients

Project description:Objective. To analyse the pathogenesis of the diseases rheumatoid arthritis (RA) and spondyloarthritis (SpA) we investigated the protein composition in synovial fluid from the patients. Methods. Fifty-six synovial fluid (SF) were analysed with non-gel based proteomics from patients with RA (32) and SpA (24). Rheumatoid factor was measured in plasma, and cell-free DNA was measured in SF. Results. Two hundred proteins were quantified in the samples. The inflammatory proteins were more abundant in the RA group, specially proteins from neutrophil granulocytes. Cell-free DNA (cfDNA) in the SF was statistically associated with proteins that are known to form neutrophil extracellular traps (NETs). Plasma C-reactive protein (CRP), was correlated to other acute phase proteins in the SF. Minimal correlation was seen between acute phase proteins and proteins in NETs. Conclusions. The results show that in the synovial cavity in vivo neutrophils form NETs. This result sustains that neutrophils from RA patients are activated and are likely to undergo NETosis resulting in SF cfDNA specific of arthritis pathology with NETosis.

Project description:Cancer-associated fibroblasts (CAF) are crucial in regulating cancer progression, yet the molecular mechanisms that determine the tumor-regulating function of CAF are poorly understood. Here, we uncover the Notch1 pathway as a molecular determinant that controls the regulatory role of CAF in melanoma growth, invasion and metastasis, and can be manipulated to reprogram and convert CAF to act as tumor suppressors. The Notch1-determined tumor-regulating function is in part mediated by Wnt-induced secreted protein 1 (WISP1). These findings reveal the Notch1âWISP1 axis as a crucial molecular determinant in governing stromal regulation of tumor progression, and establish this axis as a potential therapeutic target Total RNA obtained from MSCD-SF transduced with Cre-GFP/lentivirus in vitro compared to MSCD-SF transduced with GFP/lentivirus. Dynamic profiling of the protein life cycle in response to pathogens (protein)

Project description:Protein expression is regulated by production and degradation of mRNAs and proteins, but their specific relationships remain unknown. We combine measurements of protein production and degradation and mRNA dynamics to build a quantitative genomic model of the differential regulation of gene expression in LPS stimulated mouse dendritic cells. Changes in mRNA abundance play a dominant role in determining most dynamic fold changes in protein levels. Conversely, the preexisting proteome of proteins performing basic cellular functions is remodeled primarily through changes in protein production or degradation, accounting for over half of the absolute change in protein molecules in the cell. Thus, the proteome is regulated by transcriptional induction of novel cellular functions and remodeling of preexisting functions through the protein life cycle.
