Comparative proteomic and transcriptomic analysis of testicular tissue of yaks with or without cryptorchidism

ABSTRACT: In this study, differential genes and proteins in cryptorchidism and normal testis were screened by high-throughput transcriptome sequencing (RNA-seq) and tmt-based proteomics techniques, and the potential factors of cryptorchidism were analyzed.

Microarray profiling of total testicular transcriptome in mouse

Project description:Gene expression during spermatogenesis is highly variable and this differential pattern is very important for the successive culmination of different stages of the process, leading to production of the male gamete. Taking the time window of first wave of spermatogenesis, we did a microarray profiling of total testicular transcriptome in mouse and found several significant patterns of variable gene expression, forming upregulated and downregulated clusters among the three stages analyzed here. Mouse testis tissue were taken at three stages, 8 days, 16 days and 24 days, during first wave of spermatogenesis for RNA extraction and hybridization on the Affymetrix microarrays. Three biological replicates per age group were included and comparisons were done as 16 days vs 8 days, 24 days vs 16 days and 24 days vs 8 days testis transcriptome profile.

Klinefelter Syndrome testicular gene expression profile by a whole genome microarray approach.

Project description:Objective: Klinefelter Syndrome (KS) is the most common sexual chromosome abnormality (47,XXY) and represents the first genetic cause of male infertility. The mechanisms leading to KS testis degeneration are still unclear and no therapy is so far available for affected patients. The present study is aimed to unravel information about molecules playing a key role in the disruption of the spermatogenesis. Design: Gene expression profiles analysis of KS azoospermic testis versus normal testis, could provide useful information about the molecular basis of the alteration of the spermatogenesis. Materials and Methods: Transcriptome analysis was performed carrying out gene expression profile by a whole genome microarray approach on testis biopsies obtained from 6 azoospermic non-mosaic KS men and from 3 controls, for a total of 12 experiments. T-test and False Discovery Rate were used to evaluate differentially expressed genes. Identified transcripts were analysed by Ingenuity Pathways Analysis software to disclose genes biological functions. Results: Data analysis revealed the differentially up- and down-expression, in KS testis versus the control ones, of 656 and 247 genes related to Endocrine system development and function, Lipid metabolism, Reproductive disease, Free radical scavenging, and Cell death. Conclusions: Take together these data show the presence of several genes involved in testis microenvironment deregulation leading to the spermatogenesis failure. This information, associated with an early diagnosis could help to unravel possible therapeutic targets for testis failure prevention and limitation.

T-test and False Discovery Rate were used to evaluate differentially expressed genes. Identified transcripts were analysed by Ingenuity Pathways Analysis software to disclose genes biological functions. Results: Data analysis revealed the differentially up- and down-expression, in mosaic KS testis versus the control ones, of respectively 303 and 747 genes related to Endocrine system disorders, Lipid metabolism, Reproductive disease, cell and organ morphology, and cell death. Conclusions: Taken together these data show the presence of several de-regulated genes involved in mosaic KS testis failure. Data show the defects in sperm production and development and defects within the somatic component of the testis. This information, associated with an early diagnosis could help to unravel possible therapeutic targets for testis failure prevention and limitation.

Identification of sex-biased miRNA involved in sex differentiation and spermatogenesis in yellow catfish by comparative transcriptome analysis

Project description:In this study, three small RNA libraries constructed from gonad tissues of XX female, XY male and YY super-male yellow catfish were sequenced by Solexa high-throughput sequencing technology to investigate the expression pattern of sex-biased microRNA. The sequencing data generated a total of 384 conserved miRNAs and 113 potential novel miRNAs, among which 23, 30 and 14 miRNAs were specifically detected in XX ovary, XY testis, and YY testis, respectively. Interestingly, more abundant piRNAs were found in ovary compared to testis in yellow catfish, which phenomenon is also observed in other fish species but opposite in mammalians. We detected a number of microRNAs differentially expressed between ovary and testis, such as miR-21, miR-462, miR-430 and -200 family. When compared the transcriptome between XY and YY testis, we observed relative lower expression of miR-141 and miR-429 in YY testis. Histological analysis indicated that YY super-males have more spermatids and less spermatocytes in spermatogenic cyst than XY males under the same age and culturing conditions. The expression level of miR-141 and 429 coincides with the progression of spermatogenesis both in yellow catfish and human. At last, The expression pattern of nine arbitrarily selected miRNAs detected by quantitative RT-PCR was consistent with the Solexa sequencing results. Our study provides a comprehensive miRNA transcriptome analysis for gonad of yellow catfish with different sex genotypes, and identifies a number of sex-biased miRNAs that are potentially involved in sex differentiation and spermatogenesis.
