Project description:Transcriptional profiling of seeds of Medicago truncatula during maturation. To identify genes that are regulated during seed maturation in the model legume Medicago truncatula, plants at flowering stage were grown at variable light and temperature conditions under greenhouse environment (period March-June). Characterizing seed weight related genes through transcriptome analyses of wild and cultivated soybeans

Project description:Cultivated soybean has domesticated in China for a long history, and there are several significant phenotypic differences between wild and cultivated soybeans. Seed of cultivar is generally larger than wild soybean, therefore here we comprehensively analyzed transcriptomes of thirteen soybean accessions seeds including seven wild soybeans and six landraces through applying strand-specific RNA sequencing. Differential expressed genes related seed weight were identified, some of them were known to be associated with seed development in Arabidopsis. Noncoding RNAs are known to play important roles in plant development, and we profiled the expression pattern of long noncoding RNA (lncRNA) in cultivated and wild soybean seeds. We have identified 1,251 long intergenic noncoding RNA, 243 intronic RNA and 81 antisense lncRNA, transcriptional levels of a number of lncRNAs were significantly different between cultivated and wild soybeans, suggesting that lncRNA may be involved in soybean seed development.

Succinylome analyses in the germinating rice seed

Project description:The dehulled rice (Oryza sativa L. japonica. cv. Nipponbare) seeds were washed with distilled water for three times and then imbibed with distilled water in dark growth chamber at 26 °C and 70% relative humidity. About 100 embryos (50 mg) of rice seeds were sliced manually and collected at intervals of 24h after imbibitions (HAI), respectively. After frozen with liquid nitrogen, the samples were stored in −80 °C until used for protein extraction.

Project description:We report the changes in transcriptome that results from RNAi suppression of 7S and 11S storage protein synthesis in soybean seeds, Proteomic results show that other proteins compensate for the storage protein shortfall. The transcriptome analysis show numerous changes result from suppressing storage proteins but compensating proteins are accumulated without parallel changes in the protein's transcription.

Genome-wide transcriptional profiling in relation to seed storage compounds in tetralocular Brassica rapa

Project description:Purpose: To elucidate the physiological and molecular mechanisms underlying seed development, we conducted a genome-wide transcriptional profiling of developing seeds of ‘Sarsyun’ at four different time points (21, 28, 35, and 42 DAF). 34,423 contigs from four different developing seeds (21, 28, 35, and 42 DAF) were analyzed for transcript abundance and changes to the timing of transcript abundance in relation to the accumulation of seed storage products. Most genes involved in seed photosynthesis and carbohydrate metabolism were highly expressed at 21 or 28 DAF and were subsequently downregulated. Expression of genes coding for oleosins and fatty acid synthesis and elongation markedly increased at 28 DAF through 35 DAF, respectively, remaining high thereafter. Expression of major storage protein genes increased at 28 or 35 DAF. Overall, our results showed that dynamic changes to transcript abundance of most genes in relation to seed storage products occurred between 28 and 35 DAF.
