Proteomic Analysis of Biofilm Formation in Aeromonas hydrophila LP-2: Impact of orf-2341 Deletion

ABSTRACT: This project focuses on the proteomic analysis of biofilm formation in Aeromonas hydrophila LP-2. By comparing the wild-type strain and the orf-2341 deletion mutant, we aim to identify biofilm-associated proteins and explore the role of orf-2341 in outer membrane protein assembly and compensatory pathways. The study utilizes advanced proteomics techniques, including LC-MS/MS, to uncover differential protein expression patterns.

Preliminary investigation into the impact of GGDEF family gene deletion on the phenotype of Aeromonas hydrophila

Project description:This study took Aeromonas hydrophila as an example to study the GGDEF family gene and explore its biological function in Aeromonas hydrophila and its influence on c-di-GMP concentration. In order to systematically study the biological function of GGDEF family genes, 36 strains (54 strains screened by whole genome sequencing) with gene deletion mutation were first constructed, and then phenotypes such as biofilm, crawling and extracellular protease activity were detected. The concentration of c-di-GMP in aeromonas hydrophila was determined by HPLC. At the same time, the deletion strains with GGDEF domain that had a significant effect on the physiological function of Aeromonas hydrophila were screened for mass spectrometry identification, so as to explore the possible molecular mechanism of GGDEF domain protein affecting the physiological function.

Transcriptomic analysis of channel catfish intestine response to Aeromonas hydrophila infection correlated to skin integrity and fed state

Project description:The study focuses on the intestinal immune responses of channel catfish (Ictalurus punctatus) under various feeding and fin-clipping conditions after infection with Aeromonas hydrophila. Transcriptomic data revealed significant activation of pathogen recognition pathways (TLR signaling and C-type lectins) and pro-inflammatory cytokines, particularly in the fed groups. Histopathological findings at 2 hours post-challenge (HPC) showed minimal to moderate expansion of the lamina propria by edema in the intestines of the treated groups. At 4 HPC, mild to moderate edema with lymphocytes and plasma cells was observed, indicating early immune cell infiltration. By 8 HPC, degenerative changes in the superficial epithelium and hemorrhage were evident, especially in the fed fin-clipped group, which correlates with increased mortality in this group. The unfed groups demonstrated delayed but sustained immune responses, marked by lower histological damage compared to the fed groups. These findings provide insights into how nutritional status and fin-clipping shape the intestinal immune response and tissue damage during infection.
