Project description:Fleshy fruit help in reproduction by developing structures that, reached a given maturity stage, become attractive to animals that, feeding on them, spread the contained seeds in the environment. According to the species, seed development can be at the same pace with the pericarp, or be ready for dispersion before or after the ripening of the fruit. In peach, seed and mesocarp maturation are on pace in mid-season cultivars, while in early ripening ones the seed is not mature at ripening, with the endosperm not yet fully reabsorbed. On the contrary, slow ripening genotypes show the opposite, i.e. a mature seed in a fruit unable to fully ripen. To gain insights on the molecular control of peach fruit development and ripening and on the interactions between the seed and the mesocarp, genome wide transcriptional changes of the two fruit parts have been investigated throughout development from flower anthesis up to commercial ripening in the mid-season cultivar Fantasia. Besides flower, six time points encompassing the four fruit developmental stages were investigated.

Project description:With the quantitative profiling of consecutive tissue growth stages, we provide a global overview of expression changes of individual genes that play a role in dynamic cellular processes and developmental changes. We used tandem mass tag-based mass spectrometry and RNA sequencing to generate proteomic, phosphoproteomic and transcriptomic expression profiles for rosette leaf, flower and fruit tissue samples of the model plant Arabidopsis thaliana. In doing so, we have generated a multi-omics dataset of progressive changes during the development of three important plant organ types that can be used to study the underlying molecular mechanisms from different angles.

Flower parts, sepal, anther, ovary and stigma were collected from 8-10 different plants at R1 stage growing under three above mentioned stress conditions, and under control conditions 10 days after initiation of the stresses. Differential gene expression compared to control was studied using RNAseq method.

Whole buds, flowers or fruits were collected at various developmental timepoints. Total RNA was extracted from 10 stages of flower and fruit development. A single technical and biological repeat was performed. cDNA was amplified and labelled using standard Affymetrix protocols.

Project description:Most developmental processes associated with fruit patterning take place at the floral meristem (FM). Age-regulated microRNA156 (miR156) and gibberellins (GA) integrate to control flowering time, but it is unclear how GA and miR156 interplay during fruit patterning. Here, we used genetic, molecular, and imaging tools to demonstrate that GA and age synergistically control tomato reproductive development. We found that low levels of miR156-targeted SQUAMOSA PROMOTER BINDING PROTEIN–LIKE (SPL/SBP) transcripts or high GA responses/levels led to enlarged FMs, defects in FM determinacy, and fruits with increased locule number. Conversely, low GA responses reduced locule number and indeterminacy, and the expression of a miR156-resistant form of SlSBP15 (rSBP15) reduces FM size by decreasing cell size and cell number. Importantly, we discovered that GA levels/responses may be partially responsible for the fruit defects observed in 156OE and rSBP15 plants. GA and miR156-targeted SlSBPs regulate classical genes associated with floral determinacy, such as CRABS CLAWa (SlCRCa), and GA and SlSBPs act synergistically with tomato CLAVATA3 (SlCLV3) to control early flower and fruit development. Our findings indicate that gibberellins and age-dependent miR156-targeted SlSBPs co-operate to regulate FM activity and locule formation, thereby offering a novel mechanism of controlling fruit patterning.

Project description:The quality of the pepper fruit is significantly influenced by the properties of its surface such as color, glossiness and texture. The fruit surface is composed of a peel containing several layers including the cuticle, epidermis and the hypodermis. The peel acts as a protective barrier against biotic and abiotic stresses and is the most critical tissue affecting water loss during post harvest storage. The peel is composed of an outer epidermis with thick waxy (lipid) cuticle and few cell layers of thick-walled hypodermal cells. Despite its agronomic importance and due to the fact that the majority of studies in fruits have been conducted using flesh and peel tissues as a whole, the biochemical and genetic bases of variation in peel properties are largely unknown. In this proposal we aim to determine peel-specific gene expression in pepper by micro array hybridizations of peel and flesh RNA extracted at different developmental stages of the fruit. The cultivar Celica (Capsicum annuum) that has a large blocky fruit will be used for studying gene expression in the peel and flesh. Plants were grown in the greenhouse during the spring of 2006. Fruits were harvested at three developmental stages: young- 10 days after anthesis, mature green- 30 days after anthesis and ripe red- 45 days after anthesis. These stages were chosen because each represents a distinct phase in fruit development. At each stage, a biological replicate consists of bulked tissue from 3 fruits from each of 3 plants (a total of 9 fruits). We have a total of 4 biological replicates. For each fruit, the peel was separated from the flesh by manual dissection using thin forceps and scalpel blade. Peel and flesh samples were immediately frozen in liquid nitrogen and stored at -800C until RNA extraction. Total RNA was extracted using the GenElute Mammalian Total RNA Miniprep kit (Sigma). Project description:During the reproductive stage, tomatoes require nighttime temperatures below 21°C and daytime temperatures below 32°C for healthy flower development and fruit set. Prolonged exposure to temperatures above the optimal range can cause abnormalities in pollen development, leading to reduced fertility or sterility. This project aims to identify alterations in proteome expression and elucidate heat tolerance mechanisms affecting pollen development under elevated temperature conditions. Two tomato varieties—the heat-tolerant ‘Black Vernissage’ and the heat-sensitive ‘Micro-Tom’—were exposed to three temperature treatments: 26 °C (control), 32 °C, and 37 °C (heat-treated). Flower buds containing meiotic pollen mother cells prior to the tetrad stage and free microspores were harvested, and single cell type samples were collected using laser microdissection. Tandem mass tag (TMT) proteomics was employed to identify differential proteomic responses in pollen mother cells and free microspores between stress-treated and non-treated plants.
