Project description:The objective of this study was to identify changes in gene expression levels between wild-type and NHE4-knockout stomach. 6 wild-type and 6 NHE4-knockout stomach RNA samples were compared. Total RNA was collected from both male and female adult (between 8 and 12 weeks of age) wild-type and NHE4-null mice on a mixed 129 Black Swiss background. With the exception of one, all comparisons were done between wild-type and NHE4-null samples from littermate age- and gender-matched siblings.

The evolution of genic piRNA cluster loci expression patterns

Project description:This study examines the conservation of Piwi-interacting RNA (piRNA) clusters that come from protein coding gene transcripts. By sequencing small RNA libraries from gonad tissues of Drosophilids and Glires we discover a diverse set of genic piRNA clusters conserved across animals. This dataset reveals new expression patterns for genic piRNA clusters and examines whether changing piRNA expression patterns correlates with sequence changes in piRNA cluster genomic sequence across a variety of animal species. We dissected gonad tissues consisting of ovaries from wildtype adult Drosophilids (D.melanogaster, D.erecta, D.yakuba, D.virilis) and testes from wildtype pre-pubsecent and adult mouse and rats. Adult testes from rabbits were purchased from Pel-Freez Biologicals. Total or immunopreciptated RNAs were extracted from the pulvirized gonad tissues of Drosophilids and Glires. Small RNAs were purified from these samples, converted into cDNA libraries, and sequenced on an Illumina HiSeq2000.

Gene expression changes in testes from Abcg8 knockout mice compared to wild-type mice or to Abcg8 knockout mice where plant sterol entry was blocked by ezetimibe

Project description:Abcg8 knockout mice, fed chow, are infertile. These mice also are susceptible to absorption and retention of xenosterols contained in the diet. Ezetimibe, a blocker of dieary sterol entry, can reverse the fertility in Abcg8 knockout mice. In order to identify if the xenosterols resulted in disruption of a specific set of genes in the testes, we compared wild-type testes, Abcg8 knockout testes, or testes from Abcg8 knockout mice fed chow supplemented with 0.005% ezetimibe. To identify genes of interest, we reasoned that any causative gene expression changes seen in Abcg8 knockout mice fed chow, resulting in xenosterol exposure and accumulation, leading to infertilty, would be reveresed back to a pattern seen in the wild-type mice, when Abcg8 knockout mice are raised on chow containing ezetimibe (which prevents xenosterol absorption in the intestine). The target of this drug, Npc1l1, is not expressed in the testes.

Alteration of renal gene expression affected by atypical chemokine receptor D6 knockout in diabetic OVE26 (OVE) mice

Project description:The D6 receptor can bind and internalize inflammatory chemokines without activating intracellular pathways. D6 can reduce tissue inflammation, presumably by scavenging inflammatory chemokines. In diabetic kidney there is extensive inflammation but the possible role of the D6 receptor has never been tested. In this study, we included the renal gene expression dataset generated from whole kidney RNA extraction in diabetic mice with or without D6 gene knockout, their controls are age and sex matched D6 deficient or wild type mice on FVB background. These data are used to analyze the effects of D6 deletion on progression of diabetic nephropathy.

Project description:Ip6k1 knockout male mice are infertile and display a delay in the first wave of spermatogenesis. To understand the underlying basis of this delay, we compared the gene expression profiles of whole testes from 17 days postpartum (17dpp) and 26 days postpartum (26dpp) Ip6k1 wild type (Ip6k1+/+) and Ip6k1 knockout (Ip6k1-/-) mice. We observed deregulation of several biological processes in Ip6k1-/- testes compared with Ip6k1+/+ testes.
