Evaluation of the safety and biocompatibility of a novel galvanized vascular stent in a rabbit atherosclerosis model

ABSTRACT: Compared to traditional pure zinc stents, the new galvanized vascular stent (GVS) combines the mechanical advantages of cobalt-chromium alloy with the biological functions of zinc. This study aims to evaluate the safety and biocompatibility of the GVS in a rabbit atherosclerosis model.

Development of gene expression signatures for progression and regression of atherosclerosis

Project description:To further study the important genes related with the diagnosis and prognosis of atherosclerosis, we have employed whole genome microarray expression profiling as a discovery platform to identify genes with the atherosclerosis. we established a rabbit model of progression and regression of atherosclerosis through balloon injury and high-cholesterol diet and normal diet. Rabbit abdominal aortas from atherosclerosis among control, model group (progression) and recovery groups (regression) were collected. We found that the differentially expressed genes mainly enriched in four clusters and genes associated with inflammation and extracellular matrix were returned to or close to normal levels much earlier than the genes associated with metabolism and sarcoplasmic proliferation, they kept down-regulation or up-regulation for a long time after normal diet. Expression of four genes (LPL，BMP7，MCP2 and SST) from this signature was quantified in the same RNA samples by real-time PCR and maybe used as novel biomarkers and targets for improving the diagnosis and prognosis of atheroscleross in the future.

Ligustilide targets SMAD3, obstructing vascular fibrosis process in preventing atherosclerosis

Project description:Atherosclerosis is a persistent inflammatory state accompanied by lipid overload. Vascular fibrosis is one of the primary causes of atherosclerosis development. Although ligustilide (Lig) was shown to exert obvious antiatherogenic effects in previous studies, its precise mechanism has not been comprehensively discussed. In this paper, pharmacologic studies were performed to explore the pharmacodynamic effects of Lig on protecting aorta vascular wall structures and modulating serum inflammatory factors in ApoE-/- mice. Chemical proteomics based on a Lig-derived photoaffinity labelling (Lig-PAL) probe were applied to identify potential therapeutic targets. Mothers against decapentaplegic homologue 3 (SMAD3), which is closely related to the development of vascular fibrosis and atherosclerosis, was identified as a potential target of Lig. Lig suppressed the phosphorylation and nuclear translocation of SMAD3 by blocking the interaction between SMAD3 and transforming growth factor-β (TGF-β) receptor 1, thereby inhibiting the collagen synthesis process, preventing vascular fibrosis and improving atherosclerosis. The quantitative proteomics results from Lig-treated atherosclerotic ApoE-/- mice also indicated that Lig inhibits the expression of collagens I and III, interferes with collagen fibril organization processes and protects the aorta from vascular fibrosis. Hence, developing a novel SMAD3 inhibitor may present another therapeutic option for preventing atherosclerosis.
