ABSTRACT: General anaesthesia, especially sevoflurane inhalation anaesthesia, is an independent risk factor for postoperative cognitive dysfunction. However, the molecular mechanism by which sevoflurane inhalation alters postoperative cognitive function remains unclear.

Comprehensive analysis of differentially expressed miRNAs in aged mice presenting with postoperative cognitive dysfunction

Project description:Postoperative cognitive dysfunction (POCD) is a common postoperative complication observed in elderly patients. However, the underlying mechanisms of POCD remain unclear. MicroRNAs (miRNAs) which are involved in the pathogenesis of neurodegenerative diseases, may also affect POCD. To identify the relevant potential mechanisms, in this study, we comprehensively compared the expression profiles of miRNAs in 5 hippocampal tissues from POCD mice and 5 hippocampal tissues from control mice by microarray. A total of 128 miRNAs, whose expression changed significantly, were identified. Four miRNAs were confirmed by quantitative real-time polymerase chain reaction (PCR) in 10 paired samples. Gene Ontology (GO) and KEGG pathway enrichment analyses were performed to explore the principal functions of the deregulated genes. We predicted the potential targets of miRNAs by using bioinformatics methods. In summary, novel miRNAs were identified as potential biomarkers and therapeutic targets for POCD treatment.

Project description:This study investigates the roles of neurons, microglia, astrocytes, and oligodendrocytes in the pathogenesis of postoperative cognitive impairment, focusing on a dysregulated glia-neuron cycle. We utilized 18-month male C57BL/6 mice to model this condition, conducting single-cell RNA sequencing in the hippocampus to explore the neuroglial interactions and their implications in neuroinflammation, synaptic dysfunction, and myelin loss. This dataset includes transcriptomic profiles aimed at decoding the cellular communication in aged hippocampal cells and assessing the impact of therapeutic interventions on postoperative cognitive decline.

Identification and functional analysis of a potential key lncRNA in aged mice with postoperative cognitive dysfunction

Project description:Postoperative cognitive decline (POCD) is emerging as a significant complication of surgery in elderly patients. However, the molecular mechanisms of POCD are elusive. Here we applied microarray analyses to identify long non-coding RNA (lncRNA) and mRNA expression profiles in hippocampal tissues from a mouse model of POCD and control mice. A total of 869 lncRNAs and 690 mRNAs were differentially expressed between POCD and control mice. Subsequently, six dysregulated lncRNAs and six mRNAs were confirmed via quantitative real-time polymerase chain reaction (qRT-PCR) in 8 paired samples. Project description:Background: Postoperative cognitive dysfunction (POCD) is a debilitating neurological complication in surgical patients. Current research has focused mainly on microglial activation, but less is known about the resultant neuronal synaptic changes. Recent studies have suggested that silent information regulator 1 (SIRT1) plays a critical role in several different neurological disorders via its involvement in microglial activation. In this study, we evaluate the effects of SIRT1 activation in a POCD mouse model. Methods: Exploratory laparotomy was performed in mice aged 12-14 months under sevoflurane anesthesia to establish our animal POCD model. Transcriptional changes in the hippocampus after anesthesia and surgery were evaluated by RNA sequencing. SIRT1 expression was verified by Western Blot. Mice were treated with SIRT1 agonist SRT1720 or vehicle after surgery. Changes in microglia morphology, microglial phagocytosis, presence of dystrophic neurites, and dendritic spine density were evaluated. Cognitive performance was evaluated using the Y maze and Morris water maze. Results: SIRT1 expression levels were downregulated in POCD. Exposure to anesthesia and surgery lead to alteration in microglia morphology, increased synaptic engulfment, dendritic spine loss, and cognitive deficits. These effects were alleviated by SRT1720 administration. Conclusion: This study suggests an important neuroprotective role for SIRT1 in POCD pathogenesis. Increasing SIRT1 function represents a promising therapeutic strategy for prevention and treatment of POCD.

CD22 Blockade Alleviates Anesthesia Surgery-Mediated Cognitive Impairment in Aged Mice

Project description:<p>Postoperative cognitive dysfunction (POCD) is a common complication characterized by cognitive decline following surgery, with neuroinflammation serving as a critical pathogenic mechanism. Emerging evidence indicates that CD22, an inhibitory receptor primarily expressed on B cells and microglia, plays a key immunomodulatory role in the central nervous system. This study aims to investigate the role of CD22 in mitigating neuroinflammation and ameliorating POCD. Using a murine model of sterile laparotomy, we observed that surgery-induced cognitive impairment was associated with exacerbated hippocampal inflammation and enhanced microglial activation. In contrast, upregulation or agonism of CD22 signaling significantly reduced the release of pro-inflammatory cytokines, including TNF-α and IL-1β, and suppressed microglial overactivation. Notably, CD22-modulated mice exhibited markedly improved performance in behavioral tests assessing learning and memory compared to control groups. Our findings demonstrate that CD22 activation confers protection against POCD by inhibiting surgery-induced neuroinflammation through the S100a9 signaling pathway. Therefore, targeting the CD22 pathway may represent a promising therapeutic strategy for the prevention or treatment of postoperative cognitive dysfunction.</p>

Low blood S-methyl-5-thioadenosine is associated with postoperative delayed neurocognitive recovery

Project description:Elderly individuals display metabolite alterations that may contribute to development of cognitive impairment following surgery and anesthesia. However, these relationships remain largely unexplored. The study aims to assess the S-methyl-5-thioadenosine (MTA) is associated with postoperative delayed neurocognitive recovery (dNCR). We assess altered metabolites following anesthesia/surgery in both mice and patients to identify blood biomarkers of dNCR. Preoperative and postoperative plasma metabolites are determined by widely targeted metabolomics. The brains of mice with anesthesia/surgery show decreased MTA and activated MTA phosphorylase. Mice also show that preoperative administration of MTA can prevent inflammation and cognitive decline. In clinical patients, we detect lower preoperative serum MTA levels in those who developed dNCR. Both low preoperative and postoperative blood MTA levels are associated with increased risk of postoperative dNCR. These results suggest that anesthesia/surgery induces cognitive decline through methionine synthesis pathways and that MTA could be a perioperative predictor of dNCR.

Identification of the potential gene regulatory networks and therapeutics in aged mice with postoperative neurocognitive disorder (miRNA)

Project description:Postoperative neurocognitive disorder (PND) is one of the most common postoperative neurological complications in aged patients. In order to detect the differential expression profiles of genes caused by PND, a total of 26 18-month-old male C57BL/6 mice were randomly assigned to control group and PND group. Behavioral tests showed that mice in the PND group had impaired cognitive function compared with the control group. Three mice in each group were randomly selected to harvest the brain for analysis the expressions of circRNAs, miRNAs and mRNAs in the prefrontal cortex by next-generation sequencing (NGS) technology. Differentially expressed genes, including 1192 circRNAs, 27 miRNAs and 266 mRNAs were identified, and its accuracy was further confirmed by qRT-PCR.
