Project description:Allergic asthma is a complex trait. Several approaches have been used to identify biomarkers involved in this disease. This study aimed at demonstrating the relevance and validity of microarrays in the definition of allergic asthma expression pattern. The authors compared the transcript expressions of bronchial biopsy of 2 different microarray experiments done 2 years apart, both including nonallergic healthy and allergic asthmatic subjects (n = 4 in each experiment). U95Av2 and U133A GeneChips detected respectively 89 and 40 differentially expressed genes. Fifty-five percent of the U133A genes were previously identified with the U95Av2 arrays. The immune signaling molecules and the proteolytic enzymes were the most preserved categories between the 2 experiments, because 3/4 of the genes identified by the U133A were also significant in the U95Av2 study for both categories. These results demonstrate the relevance of microarray experiments using bronchial tissues in allergic asthma. The comparison of these GeneChip studies suggests that earlier microarray results are as relevant as actual ones to target new genes of interest, particularly in function categories linked to the studied disease. Methods: This study established an ovalbumin (OVA)-induced mouse model of allergic asthma to explore the anti-allergic effects and potential molecular mechanisms of ginsenoside Rg3. Project description:This study aims to elucidate the molecular mechanisms underlying the therapeutic effects of mountain-grown ginseng in allergic asthma and to systematically identify and characterize bioactive monomeric compounds responsible for these effects. We found that total saponins from mountain-grown ginseng significantly alleviated asthmatic phenotypes. Further multi-omics analyses revealed that these effects may be mediated through modulation of the arachidonic acid metabolic pathway, particularly by regulating the enzymatic activity of the rate-limiting enzyme PTGS2, thereby suppressing the production of PGE2. Subsequent component identification using high-performance liquid chromatography, combined with computational modeling, identified ginsenoside Rd2 as a key bioactive compound. In vitro cellular experiments, together with site-directed mutagenesis of the predicted binding sites, further confirmed that Rd2 exerts inhibitory effects on PTGS2 enzymatic activity through specific molecular interactions. Collectively, these findings provide mechanistic insights into the anti-asthmatic effects of mountain-grown ginseng and highlight Rd2 as a promising bioactive compound targeting PTGS2, offering potential therapeutic value for allergic asthma.

Project description:Obesity is associated with severe, difficult to control asthma, and increased airway oxidative stress. Mitochondrial reactive oxygen species (mROS) are an important source of oxidative stress leading us to hypothesize that targeting mROS in obese allergic asthma might be an effective treatment strategy. Using a mouse model of house dust mite (HDM) induced allergic airway disease in mice fed a low- (LFD) or high-fat diet (HFD), and the mitochondrial antioxidant MitoQuinone (MitoQ); we investigated the effects of obesity and mROS on airway inflammation, remodelling and airway hyperreactivity (AHR). HDM induces airway inflammation, remodelling and hyperreactivity in both lean and obese mice. Obese allergic mice showed increased lung tissue eotaxin levels, airway tissue eosinophilia and AHR when compared to lean allergic mice. MitoQ reduced markers of airway inflammation, remodelling and hyperreactivity in both lean and obese allergic mice, and tissue eosinophilia in obeseHDM mice. mROS regulates cell signalling by protein oxidation of multiple downstream targets: MitoQ reduced HDM-induced cysteine-sulfenylation of several proteins including those involved in the unfolded protein response (UPR). In summary, mROS mediates the development of allergic airway disease and hence MitoQ might be effective for the treatment for asthma, and specific features of obese asthma.

Phospholipase A2s (PLA2s), which release fatty acids and lysophospholipids from membrane phospholipids, have been implicated in exacerbating asthma by generating pro-asthmatic lipid mediators, but an understanding of the association between individual PLA2 subtypes and asthma is still incomplete. Here, we show that group III secreted PLA2 (sPLA2-III) plays an ameliorating, rather than aggravating, role in asthma pathology. In both mouse and human lungs, sPLA2-III was expressed in bronchial epithelial cells and decreased during the asthmatic response. In an ovalbumin (OVA)-induced asthma model, Pla2g3-/- mice exhibited enhanced airway hyperresponsiveness, eosinophilia, OVA-specific IgE production, and type 2 cytokine expression as compared to Pla2g3+/+ mice. Lipidomics analysis showed that the pulmonary levels of several lysophospholipids, including lysophosphatidylcholine, lysophosphatidylethanolamine, and lysophosphatidic acid (LPA), were decreased in OVA-challenged Pla2g3-/- mice relative to Pla2g3+/+ mice. LPA receptor 2 agonists suppressed thymic stromal lymphopoietin expression in bronchial epithelial cells and reversed airway hyperresponsiveness and eosinophilia in Pla2g3-/- mice, suggesting that sPLA2-III negatively regulates allergen-induced asthma at least by producing LPA. Thus, the activation of the sPLA2-III-LPA pathway may be a new therapeutic target for allergic asthma. Project description:Kimchi is a traditional Korean food widely recognized for its probiotic properties and potential health benefits. Several lactic acid bacteria (LAB) from Kimchi exhibit immunomodulatory properties, and their efficacy has been evaluated for various immune-related diseases. However, the mechanisms underlying the immunomodulatory effects of LAB are not yet fully understood. In this study, we demonstrated the immunomodulatory effects of Latilactobacillus sakei Wikim0185, isolated from sweet potato Kimchi, in an ovalbumin (OVA)-induced allergic asthma mouse model by inducing tolerogenic dendritic cells (DCs) and regulatory T cells (Tregs). Bone marrow-derived dendritic cells (BMDCs) and OVA-peptide-stimulated splenocytes isolated from OT-II mice co-cultured with Wikim0185 exhibited increased secretion of the anti-inflammatory cytokine IL-10. Oral administration of Wikim0185 in allergic asthma experimental mice alleviated symptoms, including airway hyperresponsiveness (AHR), leukocyte infiltration, and reduced Th2-type cytokine levels in bronchoalveolar lavage (BAL) fluid. Notably, Wikim0185-treated mice displayed an increased proportion of Foxp3+ Tregs in mediastinal lymph nodes. Additionally, mediastinal lymph node cells restimulated with OVA exhibited decreased secretion of Th2-type cytokines while showing increased IL-10 production. Interestingly, RNA sequencing and chromatin immunoprecipitation (ChIP)-qPCR analysis revealed that Wikim0185 induced tolerogenic DCs through epigenetic histone modifications, increasing active chromatin marks (H3K4me3, H3K9ac, and H3K27ac) on the promoter regions of tolerogenic marker genes (Pdl1, Il10, Socs1, and Socs3). These findings suggest that Wikim0185 modulates immune responses by epigenetically reprogramming DCs, thereby promoting Treg differentiation and suppressing excessive Th2 immune responses in an allergic asthma mouse model.
