Project description:Currently, non-albicans Candida species, including C. tropicalis, C. glabrata and C. parapsilosis, are becoming an increasing epidemiological threat, predominantly due to the distinct collection of virulence mechanisms, as well as emerging resistance to the antifungal drugs typically used in the treatment of candidiasis. They can produce biofilms that release extracellular vesicles (EVs) ‒ nanometric spherical structures surrounded by a lipid bilayer, transporting diversified biologically active cargo, that may be involved in intercellular communication, biofilm matrix production, and interaction with host.

Different protein cargo of extracellular vesicles from yeast or hyphal Candida albicans cells

Project description:Extracellular vesicles (EVs) are shed from cells of almost every type of cell or organism. These nano- to micrometer scaled lipid bilayer delimited spheres shuttle a set of bioactive molecules involved in many processes including cell-cell communication, host-pathogen interactions and even the sharing of microbial community resources. On the other hand, the yeast-to hypha transition is the most studied morphology transition in C. albicans and one of the most important traits of C. albicans to invade host tissue during infection. In this context, we have undergone the isolation and protein cargo characterization of EVs secreted by both major C. albicans morphologies by means, LC- MS analysis. A total of 2546 different proteins were identified from hyphal EVs in the 3 biological replicates carried out dropping this number to only 414 proteins identified in the 3 biological replicates from yeast EVs. Comparing the identified proteins of both EVs, 241 proteins were common to both type of vesicles. In terms of biological significance, we only took into consideration proteins that were identified in at least two biological replicates with at least two peptides and a q-value< 0.01.

This data set is the first label-free quantitative comparative study of the proteins in C. albicans EVs and their parent planktonic (yeast-form) cells. We identified proteins enriched in EVs versus WCL by comparing their LFQ intensities across all samples. After linking these results with EV enrichment data from other strains, we proposed a suite of putative EV marker proteins which were useful for multiple C. albicans strains and morphologies.

Impact of Group Psychological Interventions on Extracellular Vesicles in People Who Had Cancer

Project description:Background: There is some evidence demonstrating the effect of psychological interventions in improvements in health biological parameters. To best of our knowledge, no study had addressed the impact of any psychological intervention on extracellular vesicles. In addition, Mindfulness-Based Cognitive Therapy (MBCT) and Emotion Focused Therapy for Cancer Recovery (EFT-CR) in the group have never been explored regarding extracellular vesicles and the effectiveness of these was not compared yet. Objectives: 1. To explore and compare the effect of MBCT and EFT-CR on biological parameters and psychological variables in distressed people who have had breast, prostate and colorectal cancer; 2. In addition, we will explore the acceptability through recruitment and retention rates of MBCT and EFT-CR in group and evaluate whether these interventions are appropriate for a larger clinical trial. Methods: The design of this study is a parallel randomized controlled trial. Participants will be randomized into MBCT, EFT-CR or usual care. Outcome measures will be assessed before, at the end of the intervention (8 weeks) and follow-ups (24 and 52 weeks from the baseline moment). Hypotheses: The researchers expected that both interventions will have an effect on extracellular vesicles and other study biomarkers as well as improvements in psychological outcomes, compared to treatment as usual (TAU) group. Regarding the comparative effectiveness, we did not have evidence to hypothesize which one of the interventions will be superior in both biological (extracellular vesicles) and psychological outcomes. Contribution for practice: The results of this preliminary study would permit to know if there are benefits of these psychological interventions on changes in extracellular vesicles and on psychological outcomes related to health. In addition, this study will permit to determine the acceptability of conducting a larger randomized controlled trial.
