NRLP6 interacting proteins of small intestinal crypts of WT mouse

ABSTRACT: GST pull-down showing interaction of NLRP6 and ANXA2. Lysed small intestinal crypts from WT mice were incubated with GST-NLRP6 recombinant protein or GST recombinant protein as control. Proteins precipitated on the beads were resolved by SDS-PAGE followed by silver staining and differential bands were cut for mass spectrometry.

Project description:Immunoprecipitation assay was performed using small intestinal crypts from WT and ERAdP-HA mice. Crypts were lysed and incubated with anti-HA antibody and protein A/G beads or only protein A/G beads as control. Proteins precipitated on the beads were resolved by SDS–PAGE followed by silver staining and differential bands were cut for mass spectrometry.

Heterologous Expression of MBP-AtABC1K6-His in E. coli

Project description:Heterologous E. coli expression of AtABC1K6 tagged with maltose binding protein (MBP) and 6xHis tags revealed a spontaneous cleavage event of the recombinant protein during purification. Resulting bands were resolved on SDS-PAGE and excised for MS/MS identification. Mass spec-based identification of the individual bands determined the cleavage event as ocurring after Lysine-443. The results demonstrate that the resulting ca. 80 kD band, showing kinase activity, comprises the AtABC1K6-6xHis fragment lacking the first 71 residues of AtABC1K6.

Phosphorylation site identifcaiton of OsHSFA4d

Project description:To identify the residues of OsHSFA4d phosphorylated by OsCDPK24, the in vitro kinase assays were conducted by incubating recombinant MBP-HA-OsCDPK24 + GST-OsHSFA4d and MBP-HA + GST-OsHSFA4d. The reaction was terminated by adding SDS loading buffer to a 1×concentration, and the reaction products were subjected to CBB staining and immunoblot analysis as described above. The CBB-stained protein bands corresponding to phosphorylated OsHSFA4d were excised and subjected to LC-MS/MS analysis.

Mass spec results of Tmc7-GST pull-down

Project description:For the GST pull-down of DRG ganglion, the GST- or Tmc7-GST-associated beads were further incubated with cell lysates derived from DRG ganglion. The pulled-down proteins were then separated by SDS-PAGE gel electrophoresis and subsequently stained with silver staining Kit. Finally, the target molecular weight bands and undenatured beads were analyzed by proteomic analysis, respectively.

Project description:Cell lines of different origin (JAR (choriocarcinoma) and THP1 (monocytes, both untreated and activated by LPS/PMA)) expressing the WD40 domain of ATG16L1 fused to GST were lysed and the resulting supernatants subjected to agarose-GSH immunoprecipitation. Co-precipitating proteins were resolved by PAGE and the specific bands excised for identification using an LTQ-Orbitrap Velos device.

In the gastrointestinal tract, RANKL is expressed in the stromal cells of Peyer's patches, and involved in the development of the specialized intestinal epithelial cells, called M cells. To identify the genes involved in M-cell development, we treated BALB/c mice with recombinant GST-RANKL. After RANKL-treatment, epithelial cells were isolated from small intestine, and used for microarray analysis.
