ERAdP-interated proteins in small intestinal crypts of ERAdP-HA mouse

ABSTRACT: Immunoprecipitation assay was performed using small intestinal crypts from WT and ERAdP-HA mice. Crypts were lysed and incubated with anti-HA antibody and protein A/G beads or only protein A/G beads as control. Proteins precipitated on the beads were resolved by SDS–PAGE followed by silver staining and differential bands were cut for mass spectrometry.

Project description:Nuclei were isolated from leaves of transgenic plants expressing HA-tagged H3.1 (HTR5) or H3.3 (HTR13). MNase treated nuclear extracts were used for immunoprecipitation with HA agarose beads (Roche). Immunoprecipitated proteins were analysed by 4-20% SDS-PAGE, silver stained, and protein bands corresponding to transgenic copy of H3 and endogenous H3 were used for trypsin and LysC digestion followed by LC-MS/MS.

Project description:To identify potential phosphorylation sites of STAT6, RAW 264.7 cells were overexpressed with STAT6-HA. 48 hours after transfection, cells were treated with 10 μM etoposide for 2 h, then lysed for immunoprecipitation using anti-HA beads. Immunoprecipitates were separated by SDS–PAGE and then Coomassie blue staining was carried out. Bands of interest were cut from the gel and then in-gel digestion was performed using sequencing grade-modified trypsin. The peptides were analyzed by liquid chromatography–tandem mass spectrometry (LC–MS/MS) with nano-LC combined with Orbitrap Q Exactive mass spectrometer

Anti-HA magnetic beads were used for affinity purification of ETV6-HA (and ETV6 control) from the nuclear and cytoplasmic fraction. Following purification on-beads digest and LC-MS/MS experiments were performed at the Proteomics platform of the CHU de Quebec Research Center, Quebec, Canada.
