Mass spectrometric identification of colorectal cancer cells

ABSTRACT: Immunoprecipitation experiments were carried out to enrich TRIM24 following the lysis of DLD1 cells. The sample was added to 100 µL of 100 mM NH4HCO3, followed by trypsin (10 ng/µL) digestion at 37°C for 17 hours. Then, the sample was centrifuged at 12,000 g for 30 min, and the supernatant was freeze-dried, desalted, and freeze-dried again. Subsequently, 15 µL of 0.1% FA solution was added to dissolve the freeze-dried peptide. After centrifugation at 12,000 g for 5 min, the supernatant was transferred to the sample vial for mass spectrometry (Q Exactive HFX) analysis. The pFind (3.1.5) database search was performed, with Oxidation (M), Acetyl (Protein N-term), and Phospho (STY) set as variable modifications, and the false discovery rate of protein identification was controlled to be less than 1%.
